Abstract

The invention discloses a nucleotide mutational site for indicating high oleic acid content of rapeseeds and relates to the field of plant molecular seed breeding. When any nucleotide mutational site exists in a certain variety (strain) of the rapeseeds, the variety (strain) has a high oleic acid content property; when two nucleotide mutational sites exist at the same time, higher oleic acid content is indicated. According to the nucleotide mutational site, one rape high oleic acid mutant is used as a material, and PCR (Polymerase Chain Reaction) amplification is carried out on a rape oleic acid desaturase gene (BnFAD2), so as to obtain two high oleic acid mutational sites in the mutant. The high oleic acid rape variety (strain) containing the mutational sites is hybridized with a common variety (strain), and the mutational sites are detected in a subsequent separation generation strain, and the rape variety (strain) with the oleic acid content which is 85 percent can be efficiently and accurately obtained.
