The invention provides a graded extraction method of soybean oil and fat, and relates to the technical field of edible oil and fat processing.The graded extraction method comprises the four steps of raw material treatment, programmed cooling crystallization, solid-liquid separation and post-extraction processing.By constructing a physical separation process based on programmed cooling crystallization, soybean oil and fat are successfully extracted, and the extraction efficiency of the soybean oil and fat is improved. Compared with the prior art, the method provided by the invention has the advantages that the conventional extensive winter working section is replaced by an accurate temperature control crystallization mode, and the use of expensive technologies such as molecular distillation and supercritical extraction or organic solvents is avoided, so that the equipment transformation cost, the subsequent energy consumption and the operation and maintenance cost are greatly reduced, and the method is suitable for industrial production. The method has the economic basis of rapid industrial popularization, the process parameters are clear and controllable, continuous and stable production from raw material pretreatment, crystallization enrichment, solid-liquid separation to product refining can be realized, efficient graded utilization of resources is realized, and the overall economic benefit of a production line is remarkably improved.

The invention discloses optimization of a Cas12i3-5M-based soybean efficient multi-gene editing system and application of the Cas12i3-5M-based soybean efficient multi-gene editing system in creation of high-oleic-acid beany-smell-free germplasm resources. The system comprises a basic gene editing vector pSoC12i-V0, a promoter in a nuclease expression module is replaced by a soybean endogenous strong expression promoter GmM4pro, a human demethylase gene hsFTO synergistic module driven by GmM8pro is introduced, a tRNA processing ribozyme RNase P is adopted in a crRNA expression module for optimization, and the gene editing vector pSoC12i-V3 is constructed. The system can be used for efficiently and synchronously editing a plurality of target genes. A soybean variety 'Dongshen-89' is taken as a receptor, crRNA is designed for five genes of GmLOX1, GmLOX2, GmLOX3, GmFAD2-1A and GmFAD2-1B, and a new germplasm with high oleic acid content and no beany flavor, which is homozygically mutated at five targets, is successfully created. The invention provides an efficient gene editing tool and a new germplasm resource for soybean directional breeding and character polymerization.

Herein provided is a new runner-type peanut variety designated ‘Georgia-22MPR’ as well as the seeds, plants, and derivatives of the new peanut variety ‘Georgia-22MPR’. Methods for producing peanut plants by crossing the new peanut variety ‘Georgia-22MPR’ with itself or another peanut variety (such as another runner-type peanut variety) and plants produced by such methods are also provided. ‘Georgia-22MPR’ is a unique runner-type peanut cultivar having a combination of high-oleic and low-linoleic fatty acid ratio, a high level of tomato spotted wilt virus (TSWV), potato leafhopper, and root-knot nematode (RKN) resistance, high total sound mature kernel (TSMK) percentage, and high yield. ‘Georgia-22MPR’ provides a medium-late maturity runner-type peanut cultivar with a flat-decumbent runner growth habit, dark green foliage, medium-large runner seed size, and tan seedcoat color.
