Embodiment 1

[0034] Example 1 Obtaining of Soybean Different Vitality Groups

[0035] 1. Test material: The soybean landrace variety Da soybean seeds were used as the test material.

[0036] 2. Test method:

[0037] (1) The seeds of the landrace soybean cultivar Rhubarb soybean with consistent sowing, field management, harvest and maturity are used as test materials, and at least 10,000 seeds are used.

[0038] (2) Before the artificial aging of the seeds, put them into an artificial climate box to balance the water content of the seeds. The balance conditions are that the relative humidity is 45%, the temperature is 25° C., and the time is 15 days, so that the water content of the treated seeds is 7.4% ± 0.1 %, if the water content of the seeds does not reach this standard, continue to balance under the same conditions.

[0039] (3) After the moisture balance of the seeds meets the requirements, they are vacuum-sealed and packaged in aluminum foil bags, and divided into several parts on a...

[0042] Example 2 Using Morphological Markers to Analyze the Genetic Integrity of Soybean

[0043] 1. Analyzing samples

[0044] The treatment population of the soybean different vigor that embodiment one obtains, with reference to the standard condition in the GB / T3543.4 crop seed inspection procedure, carries out the germination test, and counts indicators such as germination potential, germination rate, germination index and vigor index (Table 1 ). Soybean parental populations with different vigor were regenerated in the field, and 200 plants were planted in each population, and the individual plants were numbered to obtain their offspring populations. 11 morphological marker traits were surveyed on 30 individual plants of each parent population during the reproductive renewal period.

[0045] After harvesting the parent populations with different vigor, individual plants are threshed to obtain the seeds of the progeny populations, and the individual plants of each populat...

[0057] Example 3 Analysis of Soybean Genetic Integrity Using SSR Molecular Markers

[0058] 1. Analyzing samples

[0059] With the analysis sample of embodiment two, different vigor parent populations and their progeny populations are during the field reproduction renewal period, get 96 individual plants (including 30 individual plants in embodiment two), collect young leaves, -80 ℃ Save it for future use. There is also a one-to-one correspondence between the single plant where the progeny collects leaves and its parent.

[0060] 2. Genomic DNA extraction

[0061] SDS method was used to extract the genomic DNA of each population of the above-mentioned parent and offspring. The extraction steps are:

[0062] (1) Put young soybean leaves into 2.0mL centrifuge tubes with tweezers, add a steel ball with a diameter of 5mm in each tube, and proceed to the next step or -80°C for later use;

[0063] (2) Quickly put the centrifuge tube into liquid nitrogen to cool down, put it int...

Abstract

The invention provides a method for analyzing genetic integrity of soybean germplasm. A method of combining a morphological marker and an SSR molecular marker is comprehensively adopted, and individual plants analyzed by virtue of the morphological marker and individual plants analyzed by virtue of the SSR molecular marker meet a relationship of corresponding one by one, namely the individual plants, subjected to agronomic trait investigation, in each population are simultaneously the individual plants undergoing SSR core primer amplification. According to the method provided by the invention, individual plants of parental population and progeny populations thereof, differing in vitality, of soybean are subjected to morphological trait investigation and are subjected to significant difference analysis; and by virtue of 20 pairs of core SSR primers, DNA amplification detect is implemented, and allele frequency and population genetic structure significant difference analysis is conducted. When differences achieve a significant or quite significant level, it regards that the genetic integrity of the germplasm is changed. The method provided by the invention is applicable to researches on the genetic integrity of the soybean germplasm; and with the application of the method, a reproductive and regenerative germination rate threshold of the soybean germplasm can be obtained more accurately.

Description

technical field [0001] The invention belongs to the field of crop germplasm resources and relates to a method for analyzing the genetic integrity of soybean germplasm. Background technique [0002] Germplasm refers to the genetic material that is directly transmitted from parent to offspring through sexual process or somatic cells and determines its inherent characteristics. For plants, the carriers carrying germplasm include not only seeds, but also plants, roots, stems, germs, cells, protoplasts, etc., and even DNA fragments. All biological types with germplasm or genes are collectively called germplasm resources (germplasm resources), also known as genetic resources (genetic resources). The diversity of genetic variation contained in crop germplasm resources is the material basis of crop breeding and biotechnology. [0003] Genetic integrity broadly refers to the state of the original genetic composition of the germplasm, that is, the genetic structure of the population...
