Embodiment 1

[0023] Example 1 Analysis of Genome-wide Expression Data in Oral Squamous Cell Carcinoma and Normal Oral Tissues

[0024] Firstly, the mRNA sequencing data of oral squamous cell carcinoma and oral normal tissues were obtained from the TCGA database, including data of 32 normal oral tissues and 341 oral squamous cell carcinoma samples. The expression level of each gene was calculated and expressed as RSEM value.

[0025] Genes that can be applied to the diagnosis of oral squamous cell carcinoma were preliminarily screened based on the following criteria:

[0026] According to the P9. Here, a total of 242 genes with high expression in oral squamous cell carcinoma were screened out. The top 30 genes are ranked according to the area under the ROC curve. Schedule 1 and figure 1 . Table 1. Expression of specific genes in oral squamous cell carcinoma and normal tissues and ROC analysis

Embodiment 2

[0035] Example 2 The expression of HOX family HOXA10, HOXA11, HOXC6 and HOXC8 was detected by pathological sections of oral squamous cell carcinoma and normal tissues.

[0036] The experimental steps are as follows:

[0037] 1. Paraffin sections (oral squamous cell carcinoma and normal tissues) were baked at 60°C for 2-3 hours

[0038] 2. Dewax with xylene twice, hydrate with gradient alcohol, and rinse with running water for 2 minutes.

[0039] 3. Use sodium citrate antigen retrieval solution, boil in microwave for 15 minutes, and cool naturally to room temperature.

[0040] 4. Add 3% hydrogen peroxide solution dropwise to remove endogenous peroxidase, incubate at room temperature for 10 minutes, wash with PBS 3 times, 5 minutes each time.

[0041] 5. Add the primary antibody at a concentration of 1:75 to each slice and incubate overnight at 4°C. The next day, rinse with PBS 3 times, 5min each time.

Embodiment 3

[0052] like Figure 5 Shown are the distribution frequencies of HOXA10(A), HOXA11(C), HOXC6(D) and HOXC8(B) gene expression values in oral normal tissues and oral squamous cell carcinoma tissues, and the TCGA database (https: / / tcga- data.nci.nih.gov / tcga / tcgaHome2.jsp) obtained level 3 expression data of oral squamous cell carcinoma, from which 341 cases of oral squamous cell carcinoma and 32 cases of normal tissues were selected, and the unit of gene expression value is RNA-Seq by expectationmaximization (RSEM).

Abstract

The invention provides a reagent kit for diagnosing oral squamous cell carcinoma. The reagent kit comprises specific antibodies for detecting HOXA10, specific antibodies for detecting HOXA11, specific antibodies for detecting HOXC6 or compositions with more than one or two optional specific antibodies for detecting the expression quantities of HOXC8. The reagent kit has the advantages that sequencing results for mRNA [messenger RNA (ribonucleic acid)] of the oral squamous cell carcinoma and normal oral tissues in TCGA (the cancer genome atlas) databases are statistically analyzed, data of 32 cases of normal oral tissues and 341 cases of oral squamous cell carcinoma tissues are incorporated, obvious difference of expression of partial genes in HOX families in the oral squamous cell carcinoma and the normal tissues is discovered, and accordingly the reagent kit is suitable to be applied to diagnosing the oral squamous cell carcinoma; expression of genes in the HOX families is detected in oral squamous cell carcinoma tissue pathological sections by the aid of immunohistochemical processes, and accordingly positive expression of the genes in the oral squamous cell carcinoma and application values of the genes in diagnosis can be determined.

Description

Technical field: [0001] The invention belongs to the field of biomedicine and relates to a diagnostic reagent, in particular to a kit for diagnosing oral squamous cell carcinoma. technical background: [0002] Oral squamous cell carcinoma is one of the ten most common cancers in the world. It is the most common oral tumor, accounting for more than 90%. There are about 500,000 new cases every year, and it is still on the rise in recent years. Oral squamous cell carcinoma not only endangers the life of patients, but also affects their beauty and appearance, as well as the quality of life of patients such as chewing, speech, and swallowing. Although there are currently comprehensive sequential therapies including surgery, radiotherapy, chemotherapy, biological therapy, and gene-targeted therapy, the long-term therapeutic effect of oral squamous cell carcinoma is still not satisfactory. According to literature reports, about 2 / 3 of the patients with oral squamous cell carcinoma...
