[0019] Further detailed explanation through specific implementation mode below:

[0020] 1. Experimental process

[0021] 1. Acquisition of human amniotic membrane: Obtained from the Zunyi Maternal and Child Health Hospital after the informed consent of the parturient and her family members. The amniotic membrane was mechanically stripped under sterile conditions, stored at 4oC, and fresh amniotic membrane was prepared within 2 hours. Prenatal serological tests were performed to exclude hepatitis B, syphilis and acquired Immunodeficiency Syndrome.

[0022] 2. Experimental animals: 72 adult healthy SD rats, weighing 180-220 g, male or female. Provided by the Experimental Animal Center of the Third Military Medical University (permit number: SCXK (Yu) 2007-0003), the animals were used in the experiment after one week of adaptive feeding in a clean animal room. The experimental process complied with the "Regulations on the Administration of Experimental Animals".

Abstract

The invention discloses a preparation method of an HAM (human amniotic membrane) neural tube and an application of the HAM neural tube in repair of injury of common peroneal nerves of peripheral nerves. The HAM neural tube for repairing injury of the peripheral nerves is prepared by mechanical exfoliation and bio-adhesion and envelops the completely transected common peroneal nerves. The number ofamniotic membrane layers constituting the amniotic membrane neural tube is determined according to the experimentally observed amniotic membrane absorption condition and function recovery time of thesutured common peroneal nerves, and satisfactory curative effects are achieved when the amniotic membrane neural tube is applied to regenerative repair of the 5mm-long common peroneal nerves after defect suture, the toe extension function, nerve conduction velocity, gross anatomy of the common peroneal nerves and histopathology of tibialis anterior muscle tissue in the damaged innervation regionare restored in a short time, the amniotic membrane is also completely absorbed within a certain period of time, uploading of peripheral stimulation signals is facilitated, axoplasm transport of injured nerve signals is also restored, expression of nerve growth factors promoting nerve axon regeneration is significantly up-regulated, and factors inhibiting axon regeneration are significantly down-regulated.

Description

technical field [0001] The invention relates to the fields of biotechnology and medicine, in particular to a preparation method and application of a human amniotic neural tube. Background technique [0002] Common clinical peripheral nerve injuries are mostly caused by trauma, which can occur alone or in combination with other tissue injuries, and can also be caused by compression, infection, ischemia, tumors, or nutritional disorders. After the peripheral nerve is injured, the movement, sensation and nutrition of the innervated area will be impaired. Clinical manifestations include pain, muscle paralysis, rough skin, no sweat, atrophy, ulceration, decreased or disappeared sensation, and blood circulation disorders. Therefore, it is necessary to restore the continuity and integrity of the nerves in time, and ensure that the nerves and myelin sheath regenerate in a suitable microenvironment, so that the damaged nerves can be repaired, the function of the damaged nerve innerv...
