Embodiment Construction

[0016] In order to make the content of the present invention easier to understand, the technical solutions of the present invention will be further described below in conjunction with specific embodiments, but the present invention is not limited to this.

[0017] 1. The florigen gene GmFT5a Analysis of expression patterns

[0018] GmFT5a The open reading frame of the gene is 519 bp in length and encodes a protein of 172 amino acids. It is a member of a subfamily of the PEBP family. For clarity GmFT2a Whether it is expressed in nodules and in different tissues at different growth stages, first analyze its expression pattern.

[0019] Soybean seeds Willimas 82 use 3% (v / v) H 2 O 2 Disinfect the surface for one minute, germinate for 7 days in quartz sand moistened with 1 / 2 total nutrient solution, and transfer to low nitrogen nutrient solution [(LN: 530μM NH 4 NO 3 +KNO 3 +Ca(NO 3 ) 2 ·4H 2 O)]. Plant growth is carried out in a long-day (LD: 14h day / 10h night) controllable greenhouse...

Abstract

The invention discloses novel application of florigen gene GmFT5a in promoting biological nitrogen fixation for leguminous plants and improving nitrogen and phosphorus nutrient efficiency. The florigen gene GmFT5a starts expressing upon rapid development of root nodules and locating at nuclei and cytoplasm of cortical cells of the root nodules, thereby significantly increasing the quantity and weight of soybean root nodules and dinitrogenase activity under both long and short durations of sunshine, increasing nitrogen and phosphorus contents in plants, and finally increasing biomass quantity of transgenic plants. Nitrogen and phosphorus nutrition in leguminous crops can be efficiently utilized; candidate gene resources are provided for high-yield molecular breeding with good photoperiod eurytopicity; the florigen gene GmFT5a has significant theoretical and practical significances in the development of environment-friendly sustainable ecological agriculture.

Description

Technical field [0001] The invention belongs to the field of biotechnology and relates to new applications of genes, in particular to florigen genes GmFT5a A new application in promoting biological nitrogen fixation of legumes. Background technique [0002] Biological nitrogen fixation refers to the process by which nitrogen-fixing microorganisms reduce molecular nitrogen in the air to ammonia to provide a nitrogen source for the growth of plants or microorganisms. In the biological nitrogen fixation system, the legume-rhizobium symbiotic nitrogen fixation accounts for more than 60% of the total biological nitrogen fixation, which is the system with the highest nitrogen fixation efficiency and the largest amount of nitrogen fixation. The formation of nitrogen-fixing nodules is a complex process of a series of physiological and biochemical changes and related gene regulation. Therefore, exploring the molecular mechanism of legume nodulation, revealing the function and molecular r...
