Abstract

The patent of the present invention discloses a microsatellite pedigree identification method of Paraloach ichthyosaurus. The method includes the acquisition of full-sib pedigrees of Paraloach ichthyosaurus; extraction of genomic DNA from parents and progeny; screening of polymorphic microsatellite markers and synthesis of primers; Fluorescent PCR amplification of DNA samples of parents and offspring; and genotype analysis of parents and offspring. The microsatellite site selected by the invention has many alleles, high polymorphism, and stable and reliable PCR products. Using these microsatellite loci to identify 17 full-sib families, the probabilities of 340 individuals finding their true parents are 95.9% and 97.9%, respectively, and the overall identification success rate is as high as 95.6%, which can meet the genetic breeding requirements of Paraloach. The identification of parental relationship and the requirements of family management provide strong technical support for the molecular breeding of Paraloach.
