The method comprises the following steps: (S1) extracting root blocks; (S2) cleaning, disinfecting and sterilizing the root blocks; (S3) transplanting the root blocks into a primary culture medium; (S4) selecting plants with adventitious buds with the lengths of 3cm-6cm, taking out the plants, and cutting root blocks, wherein each cut root block is provided with 2-3 adventitious buds; (S5) transplanting each cut root block into a proliferation culture medium, and carrying out culturing; (S6) carrying outtwo-step cutting on 7-9 plants with the adventitious bus with the length of over 5cm on the same root block: firstly cutting branches with the bud length of over 5cm, and secondly cutting the root blocks; (S7) taking out the plants, singly separating buds with the stem thickness of 1.5mm-2.0mm together with base rhizome tissues, and planting the separated plants into a rooting culture medium, andcarrying out culturing; and (S8) carrying out rooting culture for 25-35 days, integrally taking out the rooting plants, transplanting the rooting plants into a seedling hardening bag, carrying out seedling hardening, and transplanting the rooting plants into a culture container.
