Embodiment 1

[0028] 1. Experimental method

[0029] 1. Materials

[0030] Lactobacillus rhamnosus CICC20255 was purchased from China Center for Industrial Culture Collection (CICC).

[0031] 2’-FL was purchased from Shanghai Xianggeng Technology Development Co., Ltd.

[0032] LPS (lipopolysaccharide) was purchased from Sigma-Aldrich (Shanghai) Trading Co., Ltd. (sigma).

[0033] 2. Culture of Caco-2 cells

[0034]Human colon adenocarcinoma cell Caco-2 was grown in high-glucose DMEM medium containing 1% double antibody, 10% serum (FBS), at 37°C, 5% CO 2 cultured in an incubator. The cell culture medium was replaced in 2-3 days, and the cells were passaged after the cells were 80% confluent.

[0035] 3. Preparation of Lactobacillus rhamnosus CICC20255 bacterial suspension

[0036] Lactobacillus rhamnosus CICC20255 was inoculated into the improved MRS broth medium, mixed evenly and cultured in a constant temperature incubator at 37°C for 12-14h, and inoculated into the improved MRS medi...

Embodiment 2

[0069] This example illustrates an infant nutritional supplement using Lactobacillus rhamnosus and 2'-FL composition, and its weight components are shown in Table 5. The Lactobacillus rhamnosus in this example is freeze-dried bacteria powder, and the total number of live bacteria of Lactobacillus rhamnosus is not less than 1×10 11 CFU / g.

[0071]

[0072] Experimental verification found that the supplement containing Lactobacillus rhamnosus and 2'-FL has the effect of repairing intestinal epithelial cell hyperpermeability and inflammatory damage.

Embodiment 3

[0074] This example illustrates a probiotic preparation using a composition of Lactobacillus rhamnosus and 2'-FL, the weight components of which are shown in Table 6 below. The Lactobacillus rhamnosus in this example is freeze-dried bacteria powder, and the total number of live bacteria of Lactobacillus rhamnosus is not less than 1×10 11 CFU / g.

[0075] Formulation of table 6 probiotic preparation

[0076]

[0077]

[0078]It has been verified by experiments that the probiotic preparation containing Lactobacillus rhamnosus and 2'-FL has the effect of repairing the high permeability and inflammatory damage of intestinal epithelial cells.

Abstract

The invention discloses application of a composition consisting of lactobacillus rhamnosus and 2'-fucosyllactos, and belongs to the technical field of health foods. The invention further provides application of the composition consisting of lactobacillus rhamnosus and 2'-fucosyllactos in preparation of medicines or health foods or foods with a function of repairing human intestinal epithelial cellinflammatory injury, and infant formula milk powder, infant nutritious supplementary, infant complementary foods and probiotic preparations containing the composition consisting of lactobacillus rhamnosus and 2'-fucosyllactose. Researches discover that 2'-fucosyllactos is utilized by the lactobacillus rhamnosus in the composition, and the effect of repairing human intestinal epithelial cell inflammatory injury can be achieved due to interaction of the lactobacillus rhamnosus and 2'-fucosyllactos. The composition is applicable to repair of human intestinal epithelial cell inflammatory injury.

Description

technical field [0001] The invention relates to the application of a composition composed of Lactobacillus rhamnosus and 2'-fucosyllactose, and belongs to the technical field of health food. Background technique [0002] Human milk is the most natural and ideal food for infants. It is rich in nutrients and bioactive components, including protein, fat, carbohydrates, minerals, vitamins, secretory immunoglobulins, and taurine. The content of each component in human milk is moderate, and the ratio is appropriate, which is beneficial to the early growth and development of infants and the subsequent healthy growth, and has the effect of promoting the development of the infant's intestinal tract, nerves, and immune systems. HMOs are the third most abundant solid components in human milk after lactose and lipids, but have low direct nutritional value because they are not easily digested. They are synthesized in the mammary gland by glycosyltransferases and consist of 5 monosacchar...
