[0015] Experimental Example 1. Preparation and Separation of Codonopsis in Campanulaceae

[0016] The steps of the preparation and separation process are as follows: (1) Take dried platycodon root or stem and leaf samples, crush them through a 200-mesh sieve, extract with methanol or ethanol to obtain the extract, and concentrate under reduced pressure to obtain the extract; (2) Dilute the above extract with water , on D101 macroporous resin adsorption column chromatography, followed by elution with water and more than 70% ethanol, collecting 70% alcohol eluted part, concentrating the collected solution under reduced pressure, and freeze-drying to obtain a crude extract rich in tangshenoside; (3 ) Pass the obtained sample through a reversed-phase ODS column, and use 5%~85% methanol aqueous solution to elute with different concentrations, collect different effluents in sections, concentrate the effluents under reduced pressure, and finally recrystallize to obtain a purity of 95....

[0019] The steps of the preparation and separation process are as follows: (1) Take the dried Codonopsis pilosula root or stem leaf sample, crush it through a 200-mesh sieve, extract it with methanol or ethanol to obtain the extract, and concentrate it under reduced pressure to obtain the extract; (2) Dilute the above-mentioned extract with water. The extract was subjected to D101 macroporous resin adsorption column chromatography, followed by elution with water and more than 70% ethanol, collecting the 70% alcohol eluted part, concentrating the collected solution under reduced pressure, and freeze-drying to obtain a crude extract rich in tangshenoside; (3) Pass the obtained sample through a reverse-phase ODS column, and elute with 5%~85% methanol aqueous solution at different concentrations, collect different effluents in sections, concentrate the effluents under reduced pressure, a...

[0021] Experimental example 3. Preparation and separation of tangshenoside in Codonopsis pilosula

[0022] The preparation and separation process steps are as follows: (1) Take the dried root sample of Codonopsis pilosula, crush it through a 200-mesh sieve, extract it with methanol or ethanol to obtain the extract, and concentrate it under reduced pressure to obtain the extract; (2) Dilute the above extract with water, and put it on D101 macroporous resin adsorption column chromatography, eluted with water and more than 70% ethanol in sequence, collected the 70% alcohol eluted part, concentrated the collected liquid under reduced pressure, and freeze-dried to obtain a crude extract rich in tangshenoside; (3) The obtained sample was passed through a reversed-phase ODS column, and eluted with 5%~85% methanol aqueous solution at different concentrations. Different effluents were collected in sections, concentrated under reduced pressure, and finally recrystallized to obtain Codono...

Abstract

The invention discloses a preparation method of lobetyolin, C20H28O8, and application of lobetyolin in the preparation of drugs to treat antagonistic cis-diamminedichloroplatinum (CDDP) induced acute kidney injury, and belongs to the field of compound preparation and isolation and pharmaceutical use development. In-vivo and in-vitro researches show that lobetyolin can effectively alleviate acute kidney injury of mice due to CDDP, including lowering BUN and CRE levels in mouse serum, lowering TNF-alpha and IL-1 beta level in serum, and inhibiting expression levels of pro-apoptotic proteins Bax and Casapase in kidney tissue. In-vitro experiments show that lobetyolin can alleviate the activity reduction of HEK293 kidney cells and cytotoxicity, increase GSH level in the kidney cells and lower the MDA content. Therefore, lobetyolin and an effective component with the same can be prepared into drugs, health products, food additives, compound preparations or the like to prevent acute kidney injury. The drugs or health products with lobetyolin may be of any of forms, such as tablets, capsules, powder injections, injections, pills, soft capsules, granules, plasters and the like.

Description

technical field [0001] The invention relates to a new application of the compound lobetyolin in the preparation of medicines and health care products for preventing acute kidney injury, and belongs to the technical field of preparation methods of traditional Chinese medicine compounds and their medical applications. Background technique [0002] Cisplatin (CDDP), whose full name is cis-1,2-dichlorodiamminoplatinum, is one of the most effective and common antineoplastic drugs in clinical use, and it is widely used in a variety of flesh and blood in the effective dose range Cancer and solid tumor treatment, including ovarian cancer, cervical cancer, head and neck cancer and non-small cell lung cancer. However, the side effects of high-dose cisplatin limit its clinical application. According to clinical trials, common side effects of cisplatin include nephrotoxicity, neurotoxicity, ototoxicity, bone marrow toxicity and electrolyte disturbance, among which nephrotoxicity is the...
