Embodiment 1

[0041] Screening and identification of embodiment 1 erythromycin-degrading bacteria

[0042]1. Domestication, isolation and purification of erythromycin-degrading bacteria

[0043] Add 10g of soil to a Erlenmeyer flask containing 100mL of inorganic salt medium, add erythromycin 50mg / L, yeast extract 0.2%, glucose 0.1%, 30°C, 120r / min shaking culture for 3 days; add 100mg / L L erythromycin, 30°C, 120r / min shaking culture for 3 days; add 100mL inorganic salt medium and 100mg / L erythromycin, 30°C, 150r / min shaking culture for 3 days; add 100mg / L erythromycin , 30°C, 120r / min shaking culture for 3 days. After 4 rounds of acclimatization, the bacterial solution was inserted into a test tube containing 5 mL of inorganic salt medium with a 10% inoculation amount, and the acclimatization was continued with the gradient pressure acclimation method, and the concentrations of erythromycin were 100, 200, and 300 , 400, 500, 600, 800, 900 and 1000 mg / L, with erythromycin as the sole carbo...

Embodiment 2

[0053] The degradation characteristic of embodiment 2 Rhizobium Ery-6 (Rhizobiumsp.Ery-6)

[0054] a. Growth curve measurement

[0055] The bacteria were inoculated into the inorganic salt liquid medium containing 1000 mg / L erythromycin, cultured at 30°C and 120 r / min, samples were taken at different times, and the OD value of the bacterial liquid was measured by spectrophotometry at a wavelength of 600 nm, and plotted growth curve, the result is as Figure 4 As shown, within 48 hours is the logarithmic growth phase of the bacteria, the growth rate of the bacteria is slightly slow at 48-72 hours, and the growth of the bacteria tends to be stable after 72 hours. The concentration did not change significantly.

[0056] b. Degradation curve

[0057] Rhizobium Ery-6 (Rhizobiumsp.Ery-6) was inoculated in an inorganic salt medium with an initial concentration of erythromycin of 100mg / Ld according to an inoculum size of 5%, at 30°C, pH7.0, 120r / min Shake the culture under the con...

Abstract

The invention relates to a rhizobium strain, namely Rhizobium sp. Ery-6). The Rhizobium sp. Ery-6 is preserved at China Center for Type Culture Collection (CCTCC) under the preservation number of CCTCC M 2019729. The invention further provides a microbial agent containing the Rhizobium sp. Ery-6 and a preparation method of the microbial agent; and moreover, the invention still further provides application of the microbial agent in erythromycin degradation. The Rhizobium sp. Ery-6 provided by the invention has very strong degradation ability on erythromycin, so that the Rhizobium sp. Ery-6 cangrow by taking the erythromycin as the sole carbon source. In 48 hours, the Rhizobium sp. Ery-6 is capable of reaching an erythromycin degradation rate up to 79.78% on erythromycin of which the concentration is 100 mg / L in an inorganic salt medium as well as an erythromycin degradation rate up to 37.3% on erythromycin of which the concentration is 1000 mg / L in an organic salt medium; and thus, theRhizobium sp. Ery-6 has a good development and application prospects.

Description

technical field [0001] The invention relates to the technical field of microorganisms, in particular to a rhizobia and its bacterial agent and application. Background technique [0002] Antibiotics are low to moderate molecular weight compounds with diverse chemical and biological properties. Antibiotics are medicines used to treat bacterial infections. Antibiotics are widely used in animals in the process of large-scale livestock and poultry farming, including tetracyclines, sulfonamides, fluoroquinolones, macrolides, aminos, etc. China produces about 210,000 tons of antibiotics every year, accounting for about 60% of the world's total, including 36 different antibiotic compounds, of which more than half of the antibiotics are used in livestock and poultry farming, and the residual antibiotics in livestock and poultry manure enter the farmland along with fertilizers . It has been reported that almost 90% of antibiotics used in livestock and poultry are used at subtherape...
