Problems solved by technology

[0002] Huntington's disease (HD) is caused by a mutation in the IT15 gene on chromosome 4, resulting in abnormal amplification of the CAG repeat sequence polyglutamine (poly-Q), resulting in an abnormally encoded mutant Huntingtin protein (mutantHuntingtin, mHTT) Aggregates, a monogenic neurodegenerative disorder for which there are currently no effective treatments [1-3]

[0038] (1) The degradation effect of polygala saponin F on EGFP-HDQ74 transfected by mouse neuroblastoma (Neuro2a) cells. The specific process is as follows.

[0039] 1. Cell culture: subculture at a ratio of 1:5 each time

[0040] (1) Aspirate and discard the culture medium, wash once with 3-10 ml 0.01M PBS;

[0041] (2) Add 3-5 ml 0.25% trypsin-EDTA to digest for 2-3 minutes;

[0042] (3) Add 3-5 ml DMEM medium containing 10% FBS to stop the digestion, and gently pipette to separate the cells;

[0043] (4) Aspirate the cell suspension in the dish into a 15ml centrifuge tube;

[0044] (5) Centrifuge (1200rpm 2min), and discard the supernatant;

[0045] (6) Add 1ml of DMEM medium containing 10% FBS to resuspend, suck out 200μl to a 10cm petri dish, plant two dishes, suck out 67ul to a 6cm petri dish, and put it in a 37°C incubator with 5% CO2 .

Abstract

The invention belongs to the technical field of medicine, and in particular relates to an application of onjisaponin F in preparation of a drug for preventing and treating huntington disease (HD). Theresearch results of the invention show that the onjisaponin F has a degrading effect on exogenous poly-Q, and has a degrading effect on endogenous huntingtin aggregates in fibroblasts of patients with different CAG numbers from a HD family, and has a degrading effect on endogenous huntingtin in iPS-directed differentiated neurons of patients from the HD family; the onjisaponin F has no effect onendogenous huntingtin in fibroblasts of normal people from the HD family, and has no effect on endogenous huntingtin in iPS-directed differentiated neurons of normal people from the HD family; and theresults prove that the onjisaponin F can enhance autophagy on fibroblasts from the HD family and the iPS-differentiated neurons, and the results show that the onjisaponin F can be used to prepare thedrug for treating and preventing the huntington disease.

Description

technical field [0001] The invention belongs to the technical field of medicine, and specifically relates to the new medical application of monomers of known medicines, in particular to the application of polygala saponin F, a monomer of traditional Chinese medicine polygala, in the preparation of medicines for preventing and treating Huntington's disease. Background technique [0002] Huntington's disease (HD) is caused by a mutation in the IT15 gene on chromosome 4, resulting in abnormal amplification of the CAG repeat sequence polyglutamine (poly-Q), resulting in an abnormally encoded mutant Huntingtin protein (mutantHuntingtin, mHTT) Aggregates, a monogenic neurodegenerative disorder for which there are currently no effective treatments [1-3] . HD patients will have uncontrollable dance-like movements and cognitive decline, as well as mental disorders such as depression, anxiety, and dementia, and eventually cannot take care of themselves, have difficulty swallowing and...
