Embodiment Construction

[0030] In order to make the object, technical solution and advantages of the present invention more clear, the present invention will be further described in detail below in conjunction with the examples. It should be understood that the specific implementations described here are only used to explain the present invention, not to limit the present invention.

[0031] figure 1 As shown, the exosome surface functionalized PEEK material realized by the present invention is prepared by the following method:

[0032] Step 1, culturing mesenchymal bone stem cells, which are obtained from bilateral humerus and femur of female SD rats;

[0033] Among them, mesenchymal stem cells were cultured in Dulbecco's modified eagle medium Dulbecco (DMEM, GIBCO) containing 10% fetal bovine serum (FBS, GIBCO) at 37°C and 5% CO 2 .

[0034] At the same time, RAW264.7 culture was carried out, RAW264.7 cells were purchased from ATCC cell bank, in 1640 medium (GIBCO) at 37 °C and 5% CO 2 to culti...

Abstract

The invention discloses an exosome surface functionalized PEEK (Polyether-Ether-Ketone) material which is prepared by the following method: step 1, culturing mesenchymal stem cells which are taken from humerus and femur on both sides of a female SD (Sprague Dawley) rat; step 2, centrifugal separation; centrifugally separating the bone mesenchymal stem cells to prepare exosomes; step three, sulfonation: immersing the PEEK into a sulfuric acid solution to enable the PEEK to form a uniform porous structure; step 4, preparing SPEEK: carrying out hydro-thermal treatment on sulfonated PEEK to remove residual sulfuric acid so as to obtain SPEEK; 5, TA-SPEEK is prepared; reacting the SPEEK with a sample under a magnetic stirring condition to obtain TA-SPEEK; and step 6, synthesizing Exo-TA-SPEEK, namely dropping the exosome from the bone mesenchymal stem cells onto TA-SPEEK, so that the Exo-TA-SPEEK can be synthesized. According to the invention, bone formation can be mediated, immune response is controlled, and effective combination with a human body is realized.

Description

technical field [0001] The invention belongs to the technical field of biological materials, in particular to an orthopedic material for orthopedic diseases and cosmetic plastic surgery. Background technique [0002] When the bones of the human body are damaged, the bones need to be repaired. Currently commonly used materials are: [0003] 1. Natural bone materials: autologous bone, allogeneic bone, xenograft bone. [0004] 2. Bioceramic materials: bioinert ceramics, bioactive ceramics, biodegradable ceramics, bioceramic composites. [0005] 3. Polymer materials: collagen, fibrin, chitin, etc. [0006] 4. Composite materials. [0007] Traditional bone substitute materials have the ability to induce osteoblasts to form bone in vitro, which can promote bone regeneration. However, some inconsistencies were found between in vitro and in vivo experiments, such as materials that favored osteogenesis in vitro could not promote bone regeneration in vivo. Traditional bone subst...
