Embodiment Construction

[0056] Referring now to the drawings, there is shown a Raman analysis apparatus, indicated generally at 100, embodying an aspect of the present invention. Apparatus 100 may comprise an optical microscope, which in a preferred embodiment is a spinning disk confocal microscope. Accordingly, device 100 may be referred to as a confocal Raman analysis device. In some embodiments, as described in more detail below, device 100 analyzes a sample by capturing images using Raman scattered light and may be referred to as a Raman imaging device or a confocal Raman imaging device. In some embodiments, device 100 may be described as a Raman spectroscopy device or a confocal Raman spectroscopy device.

[0057] The device 100 comprises a station 20 for receiving an object 55 to be analyzed. Object 55 includes a sample, typically a biological, semiconductor, crystalline, or chemical sample, and may be located in or on a glass slide or other support (not shown). The object 55 is located in t...

Abstract

A Raman analysis apparatus includes an imaging optical system for imaging an object along an optical path to an optical detector, an irradiation optical system for irradiating the object, and a scanning device including a rotating pinhole disc. The irradiation optical system guides light onto the scanning device to irradiate the object at a plurality of irradiation points. The imaging optical system images Raman scattering light emitted from the object at the irradiation point to an intermediate image plane where the scanning device is located. The imaging optical system may include a spectral filter and / or at least one spectral analyzer. The device is capable of performing high speed confocal Raman imaging and / or high speed confocal Raman spectroscopy.

Description

technical field [0001] The present invention relates to Raman analysis, including Raman spectroscopy. The invention particularly relates to confocal Raman analysis. Background technique [0002] Confocal Raman spectroscopy using lasers and confocal microscopy is known. It involves directing a laser through a single pinhole, illuminating the sample with a single laser spot, and collecting the Raman scattered light generated by the sample through the pinhole. The collected Raman light is sent to a spectrometer, which performs a single-point Raman spectral acquisition. If Raman images are required, a motorized XY or XYZ stage is typically used to move the sample relative to the static laser spot. Alternatively, the laser can be scanned relative to the sample. In any case, images were constructed from Raman spectra acquired individually from the respective sampling points, which were illuminated one at a time. The spectrum acquired for each point provides image data represe...
