Embodiment 1

[0009] ①Take 0.5kg Artemisia annua plants (produced in Tianjin area, wild, collected in July or August, the content of artemisinin in the plants is 5.0377mg / g), use a pulverizer to crush, sieve and dry, and the crushed particle size is 60 mesh . ②Add 30 liters of petroleum ether (30-60°C) and soak at 35°C for 4 hours. ④Stir and extract at a constant temperature of 50°C for 180min, with a stirring speed of 800r / min. ⑤ After the extraction, cool the extract to room temperature and filter it with suction. ⑥Wash the filtrate with 2% NaOH solution alkali and discard the lower alkali solution, wash with distilled water until neutral. ⑦The extract after washing with water was rotatively evaporated at 50°C to obtain about 3.5 grams of artemisinin extract, and the solvent was recycled. At this time, the extraction rate of artemisinin in Artemisia annua can reach more than 90%.

Embodiment 2

[0011] ①Take 0.5kg Artemisia annua plants (produced in Tianjin area, wild, collected in July or August, the content of artemisinin in the plants is 5.0377mg / g), use a pulverizer to crush, sieve and dry, and the crushed particle size is 80 mesh . ②Add 30 liters of petroleum ether (30-60°C) and soak at 30°C for 3 hours. ④Stir and extract at a constant temperature of 52°C for 150min, with a stirring speed of 1000r / min. ⑤ After the extraction, cool the extract to room temperature and filter it with suction. ⑥Wash the filtrate with 2.5% NaOH solution alkali and discard the lower alkali solution, wash with distilled water until neutral. ⑦The extract after washing with water was rotatively evaporated at 50°C to obtain about 3.5 grams of artemisinin extract, and the solvent was recycled. At this time, the extraction rate of artemisinin in Artemisia annua can reach more than 90%.

Embodiment 3

[0013] ①Take 0.5kg Artemisia annua plants (produced in Tianjin area, wild, collected in July or August, the content of artemisinin in the plants is 5.0377mg / g), crushed with a pulverizer, sieved and dried, and the crushed particle size is 70 mesh . ②Add 30 liters of petroleum ether (30-60°C) and soak at 25°C for 4 hours. ④Stir and extract at a constant temperature of 48°C for 200min, with a stirring speed of 800r / min. ⑤ After the extraction, cool the extract to room temperature and filter it with suction. ⑥Wash the filtrate with 1.8% NaOH solution alkali and discard the lower alkali solution, wash with distilled water until neutral. ⑦The extract after washing with water was rotary evaporated at 48°C to obtain about 3.5 grams of artemisinin extract, and the solvent was recycled. At this time, the extraction rate of artemisinin in Artemisia annua can reach more than 90%.

Abstract

The present invention belongs to the field of extraction technology of chemical components in natural products. The process of extracting an separating artemisinin from sweet wormwood plant includes the following steps: crushing sweet wormwood stem and leaf, 60-80 mesh sieving and drying at 40-60 deg.c; soaking in 30-60 deg.c petroleum ether fraction at 20-35 deg.c for 3-4 hr; heating at 48-52 deg.c and under stirring to obtain extract liquid; cooling the extract liquid to room temperature and filtering; alkali washing the filtrate with 1.8-2.5 wt% concentration sodium hydroxide solution, eliminating the lower layer of alkali liquid, washing with deionized water to neutrality; rotation evaporating the washed extract liquid at 48-50 deg.c to obtain artemisinin extractum and recovering the solvent for reuse. The present invention has the advantages of simple technological process, low production cost, and high artemisinin extracting rate up to 90 %.

Description

technical field [0001] The invention relates to a method for extracting and separating artemisinin from Artemisia annua plants, which belongs to the extraction technology of natural product chemical components. Background technique [0002] Artemisinin is a colorless needle-like crystal extracted from the compound inflorescence plant Artemisia annua L., with the molecular formula C 5 h 22 o 5 , is a new type of sesquiterpene lactone, which has a peroxy bond and a δ-lactone ring, and has a 1,2,4-tri-erne structural unit including peroxides, and the molecule contains 7 chiral center. Due to its unique structure, special antimalarial mechanism, special effects against chloroquine falciparum malaria and cerebral malaria, and high efficiency and low toxicity, it is called "the only effective malaria treatment drug in the world" by WHO. At the same time, scholars at home and abroad have conducted a large number of pharmacological and clinical studies. Experiments have shown th...
