Problems solved by technology

[0004] 2. In the calcification stage of bilirubin in the above-mentioned production process, bilirubin is often lost to varying degrees due to incomplete calcification. According to calculation, according to the above calcification process, the percentage of bilirubin loss reaches 20%~30%, or even higher
[0005] Due to the above two reasons, the yield of bilirubin in the existing indirect method is relatively low, generally between 0.003% and 0.005%, and the highest can only reach Around 0.008%

Embodiment 1

[0020] 1. Reduction of biliverdin in bovine bile

[0021] Select fresh ox bile, add 2 to 3 ml / liter of reducing agent in the bile, the reducing agent adopts vitamin C with a concentration of 3 to 5%, adjust the pH value to 3.5 to 6 with sodium hydroxide or hydrochloric acid, and the temperature is 20~30℃, then let it stand for 2 hours, and prepare the liquid for later use;

[0022] 2. Formation of bile calcium salts

[0023] Put the feed liquid obtained in the previous step in a stainless steel jacketed pot (the interlayer can be heated by steam or cold water for cooling), and after adjusting the pH value to 10-12 with 30% sodium hydroxide, the liquid is mixed according to the weight ratio of the feed liquid. Add calcium sulfate to 3-7% of the solution, and slowly heat to 60-65°C. When brown-red bile calcium salt precipitates, readjust the pH value to 10-12, continue heating to boiling, keep warm for 5 minutes, and filter get bile calcium salt;

Embodiment 2

[0028] 1. Reduction of biliverdin in bovine bile

[0029] Select frozen ox bile for use, add reducing agent 4~5 ml / liter in this ox bile, described reducing agent is sodium borohydride, sodium sulfite or sodium bisulfite of 4~5% concentration, with sodium hydroxide or Hydrochloric acid is used to adjust the pH value to 5-7.5, the temperature is 40-50°C, and then stand still for 1-2 hours, and the feed liquid is set aside;

[0030] 2. Formation of bile calcium salts

[0031] Put the feed liquid obtained in the previous step in a stainless steel pot, adjust the pH value to 10-12 with 30% sodium hydroxide, add calcium chloride according to the weight ratio of the feed liquid to 2-4%, and slowly heat to 60-65°C, when brown-red calcium bile salt precipitates to form, readjust the pH value to 10-12, continue heating to boiling, keep warm for 3 minutes, and filter to obtain bile calcium salt;

Abstract

The invention provides a method for extracting bilirubin from oxgall. The method comprises three steps of reducing biliverdin in the oxgall, forming calcium bilirubinate, extracting and refining the bilirubin. The method has an improvement point of adding the biliverdin reducing step capable of reducing the biliverdin contained in the oxgall and the biliverdin oxidized during collection or storage into the bilirubin so as to increase the bilirubin content of the oxgall, which is an important step for improving yield. Meanwhile, neutral calcium chloride or calcium sulfate is added in the step of forming the calcium bilirubinate as a calcium bilirubinate forming agent, the pH value of feed liquid can not be changed when calcium ions are formed after decomposition, so as to ensure complete formation of the calcium bilirubinate in the feed liquid and improve yield. Through the improvements, the yield of the method can reach 0.008 to 0.011 percent, which is improved by 0.003 percent than the prior art.

Description

technical field [0001] The invention relates to a method for extracting bilirubin from animal bile, in particular to a method for extracting bilirubin from ox bile. Background technique [0002] As the main component of artificial bezoar, bilirubin is widely used in medicine. The extraction of bilirubin is mainly extracted from animal bile. At present, there are more and more methods for extracting bilirubin, but they are generally classified into two categories, one is direct method, also known as one-step method, such as Chinese patent 97125601.2 "Technology for extracting bilirubin by one-step method"; the other is indirect method, Also known as the two-step method, such as the "production process for extracting bilirubin from ox bile" published in Chinese patent 88105159.4, the "extracting bilirubin from ox bile" with the calcium salt method provided in the Chinese Journal of Medicine 1990.21 (8). These two types of methods have their own advantages and disadvantages, ...
