Embodiment

[0016] Add 50 mg of layered hydroxyapatite powder with a layer spacing of 3.1 nm into a conical flask (see CN101058416 for the preparation method), and add a small amount of sterilized high-purity water. Intermittently ultrasonicated 3 times, 0.5min each time, ultrasonic power 100W, ultrasonic frequency: 40KHz, and finally a uniformly dispersed hydroxyapatite suspension was obtained. Solid salmon sperm DNA (commercially available) was dissolved in 100 mL diethyl pyrocarbonate solution to prepare a solution with a concentration of 1 mg / mL. Add 1 mg of DNA solution dropwise into the Erlenmeyer flask, oscillate and sonicate, sonication conditions: intermittent sonication twice, 0.5min each time, sonication power: 100W, sonication frequency: 40KHz. Finally, adjust the volume to 20mL. Put the Erlenmeyer flask into a constant temperature shaker to vibrate, rotating speed: 150rpm, temperature: 25°C, every 0.3h of shaking, equilibrium adsorption for 1h, repeat the operation 6 times. ...

Abstract

The invention relates to a DNA and nano lamellar hydroxylapatite compound as well as a preparation method thereof. Lamellar hydroxylapatite and salmon sperm DNA are used as raw materials to be mixed in percentage by mass of 1:5-100 of DNA and lamellar hydroxylapatite. The preparation method comprises the following steps: intermittently shaking the suspension of lamellar hydroxylapatite and DNA solution at 37 DEG C to react for 5-7 times and drying at normal temperature. The compound has an interlayer type structure and the layer distance of 3.3-4.8nm and realizes the storage, protection and release of DNA with a superhelix structure. The preparation method of the compound is relatively easy and significant for applying the hydroxylapatite in aspects of polygene medicine treatment and protein / gene composite medicine treatment and has low cost.

Description

technical field [0001] The invention relates to a composite of DNA and nano-layered hydroxyapatite and its preparation method and application, in particular to the preparation of a DNA / layered hydroxyapatite nano-gene drug carrier material. Background technique [0002] Vectors that mediate gene transfer can be divided into two categories: viral vectors and non-viral vectors. Potential mutagenic and carcinogenic risks of viral vectors limit their application in gene therapy. Non-viral vectors have the advantages of relatively low toxicity, no immunogenicity and tumorigenicity, and large DNA carrying capacity. The purpose of the present invention is to establish a non-viral vector gene transfer system. Layered inorganic carrier materials for DNA intercalation have been extensively studied in recent years. DNA intercalation composite materials such as DNA / montmorillonite, DNA / layered dihydroxy compound, DNA / phyllosilicate, etc., which have emerged so far have successfully s...
