specific Embodiment approach 1

[0013] Specific embodiment one: Utilize cellulose acetate / polypropylene composite enzyme film immobilization cellulase and alkaline protease, extract soybean oil with solvent leaching method and realize through the following steps:

[0014] Step 1: Preparation of cellulose acetate / polypropylene composite membrane: Dissolve 5g of cellulose acetate in 100mL of acetone. After dissolving into a uniform membrane liquid, take the polypropylene membrane prepared before the experiment and soak it in the dissolved cellulose acetate Naturally form a film in the acetone solution of the element, rinse the cellulose acetate / polypropylene composite film with deionized water several times and dry it for later use;

[0015] Step 2: Enzyme immobilization: mix a certain amount of cellulase and buffer under its optimum conditions, add cellulose acetate / polypropylene composite membrane, stir and adsorb for a certain period of time, take out the enzyme membrane and wrap it on a cylinder Immobilize...

Abstract

The invention provides a novel oil extraction method which combines enzymolysis with a solvent and aims at overcoming the defects that lixiviation time of a lixiviation method is relatively long, the dosage of enzyme of an aqueous enzymatic method is high, and the enzyme cannot be used repeatedly. The novel oil extraction method comprises the steps that cellulase and alkali protease are immobilized under an optimum immobilization condition, and an enzyme membrane is taken out to be wound on the surface of a cylinder, so as to obtain an immobilized cellulase roll and an immobilized alkali protease roll; then the moisture of germ flakes is adjusted by adjusting the humidity of the air under a vacuum condition, and the optimum relative humidity is 75%; the immobilized cellulase roll is used to roll the germ flakes continuously for 30min to allow the cellulase to perform the enzymolysis on plant cell walls; and then the immobilized alkali protease roll is used to roll the germ flakes continuously for 30min for separating protein from oil. After the enzymolysis of the germ flakes is finished, the solvent is used to extract the oil, the extraction time is only required to be 35min, and the residual oil reaches below 0.5%. In addition, an immobilized enzyme column can act on the material germ continuously for more than 24h, and the enzymic activity is still kept at 80%.

Description

technical field [0001] The invention relates to a soybean oil extraction method, relates to the application of enzyme immobilization technology to extract soybean oil, and belongs to the field of vegetable oil extraction and processing. Background technique [0002] Soybean is an important oil crop. Soybean contains 20% oil, 40% protein and 25% carbohydrates and other nutrients. So soybean is not only an important oil crop, but also a resource of high-quality protein. At present, the main methods of extracting oil from soybeans are pressing and leaching. Although these methods have a high oil yield, they take a long time and cause protein denaturation during the oil extraction process, which reduces the efficiency of the cake after oil extraction. Aqueous enzymatic technology overcomes the economic, environmental, and safety problems of the traditional oil production process, but the amount of enzyme used is large, the price is expensive, and it cannot be reused. In additio...
