Embodiment 1

[0020] 1. Test strains: two blast fungus strains YNC08-30 and YNC08-20 collected from rice planting areas in Yunnan Province.

[0021] 2. Planting of rice: 3 kinds of wild rice (ordinary wild rice, medicinal wild rice, and verrucous wild rice) and the susceptible control variety Lijiang Xintuan Heigu (LTH). LTH was germinated in an incubator at 30°C for 3 days , sow in plastic seedling boxes (15×5×10cm), the soil is sterilized soil, sow 5 seeds per row, place them in a greenhouse at 24-30°C, topdress 0.2-0.3g of urea per box, and 3 kinds of wild Rice is a regenerated plant of perennial root, which is kept in the greenhouse.

[0022] 3. Cultivation and sporulation of rice blast strains: Cultivate the bacteria on rice flour agar medium. When the mycelia cover the surface of the medium, wash off the aerial mycelium with tap water, and place it under a fluorescent lamp for about 40 hours to promote its sporulation. Wash the spores with distilled water to make a spore suspension (...

Abstract

The invention discloses a method for identifying a rice blast-resistant gene of wild rice. The method comprises the steps of cutting middle parts of leaves of wild rice plants, and placing in culture dishes with single-layer filter paper; adding an appropriate amount of sterilized water to each dish; dropping 5mu L spore suspension on each leaf; inoculating each leaf for 5 points; covering lids; placing the culture dishes in an illumination incubator at 23 DEG C for dark culture for 24 hours; carrying out illumination culture with illumination intensity of 3400Lux / 1310Lux (12 hours / 12 hours); and investigating disease states after culturing for 6-7 days, wherein an investigation disease is divided into five levels, 0-2 level is disease-resistant, and 3-4 level is infected. According to the method, a method for inoculating rice blast is optimized; the simpler and large-scale method for identifying the resistance of the wild rice is founded; and the problem that the disease resistance is difficult to identify due to the fact that the wild rice is high in sterility rate, grains fall easily, seeds are difficult to collect, the degree of dormancy is different, and germination growth is inconsistent is solved.

Description

technical field [0001] The invention relates to a gene identification method, in particular to a method for identification of wild rice blast resistance gene. Background technique [0002] Rice blast is one of the most important diseases of rice globally, which can cause 11%-30% yield loss to rice, and the loss of rice is as high as hundreds of millions of kilograms, and the global annual loss is about 157 million US dollars. Practice has proved that the use of resistant varieties is one of the most economical and effective measures to prevent and control rice blast, and it also meets the requirements of human green food. However, due to the diversity and variability of the physiological races of Magnaporthe grisea, after 3-5 years of continuous use of resistant varieties, the resistance of the varieties will obviously decline. Therefore, finding and screening stable and long-lasting disease-resistant varieties, especially the discovery and utilization of long-lasting resis...
