Problems solved by technology

Due to the complex sample pretreatment of these analytical methods, the required instruments are expensive, the detection cost is high, and the analysis speed is slow, it is not suitable for on-site monitoring and screening of a large number of samples.
Enzyme-linked immunoassay (ELISA) has strong specificity, high sensitivity, low price, quick and easy, can make up for the shortcomings of traditional chromatographic analysis methods, and has been widely used in the detection of organic pollutants in the environment. The ELISA detection method of A has not been reported yet

Abstract

The invention discloses an indirect competition ELISA method for tetrabromo bisphenol A. The assay method comprises the following methods: 1) synthesizing an artificial antigen through the tetrabromo bisphenol A; 2) preparing a tetrabromo bisphenol A polyclonal antibody: taking the conjugate of the tetrabromo bisphenol A and BSA as an immunizing antigen, immunizing Kunming white rats, and utilizing the conventional polyclonal antibody preparation and purification technologies to acquire the tetrabromo bisphenol A polyclonal antibody; 3) assaying the indirect competition ELISA: choosing an envelope antigen of which the concentration is 10 [mu]g / mL, and performing dilution with a carbonate buffer solution; washing an ELISA plate, adding the buffer solution to seal the ELISA plate and washing the ELISA plate again; carrying out competition, washing the ELISA plate, adding a second antibody and washing the ELISA plate again; performing a chromogenic reaction; finally, measuring with a microplate reader to acquire that a light absorption value is 450nm (OD 450). The invention constructs the indirect competition ELISA method for the tetrabromo bisphenol A, and provides a quick and efficient assay means for assaying the residual tetrabromo bisphenol A in atmosphere; as the method adopts the polyclonal antibody, the cost is lower and the stability is better.

Description

technical field [0001] The invention relates to a detection method for tetrabromobisphenol A content, more specifically an indirect competitive ELISA detection method for tetrabromobisphenol A, which belongs to the technical field of immune detection. Background technique [0002] Tetrabromobisphenol A (TBBPA) is currently the most commonly used brominated flame retardant, and is widely used as a reactive flame retardant to manufacture brominated epoxy resins and brominated polycarbonate and as an intermediate to synthesize other Complicated flame retardants are also used as additive flame retardants in ABS, HIPS, unsaturated polyester, rigid polyurethane foam, adhesives and coatings. Tetrabromobisphenol A can be released from products into environmental media, and with its extensive use, the content of tetrabromobisphenol A in the natural environment is increasing rapidly. At present, TBBPA has been detected in the atmosphere, water, soil, and organisms. Studies have show...
