Problems solved by technology

Due to the long-term and large amount of foreign imports costing hundreds of millions of dollars in foreign exchange every year, not only the cost of bulbs is high, but some varieties are also of poor quality due to the difficulty in introducing seed sources, and the species are degraded, which cannot meet the requirements for export to participate in market competition.
In addition, the procedures fo...

Abstract

The invention discloses a tissue culture method for fast propagation of a new Lilium oriental hybrid variety, and in the method, a low concentration hormone formula test-tube plantlet breeding method is used for fast propagation, and river sand or silt are used as a matrix for hardening seedling culture. The method particularly includes the following steps: selecting plants of the new Lilium oriental hybrid variety for virus detection, and taking a bulb of an identified virus-free plant as an explant for explant sterilization; respectively performing primary culture, subculture and rooting culture on the sterilized explants; performing low temperature treatment before hardening seedling; selecting the river sand or the silt as the matrix for hardening seedling; and after completion of the hardening seedling, transplanting the bulb to a high altitude field for bulb seeding. According to the tissue culture method, a large number of high-quality and stable Lilium oriental seedlings can be obtained, and the shortcomings in the prior art can be overcome.

Example Embodiment

[0021] Example 1:
[0022] The technical solution of this embodiment includes the following steps:
[0023] 1) Sterilization and cultivation of explants: The self-cultivating Oriental lily hybrid varieties Zhuzhouhong, Huaxianzi, Pleasant Goat, and Lona are used as materials, and 5-10 strains of each variety are selected for virus detection, and virus-free plants are identified. Take the bulb as an explant. Explant sterilization: peel off the scales, rinse with tap water, dry, and then soak in 75% alcohol for 30 seconds, then sterilize with 0.1% mercury liters for 15-20 minutes, and rinse with sterile water 3 to 4 times. The formula of the primary culture medium is: MS+0.3~0.5mg./L 6-BA, sucrose 3%;
[0024] 2) Subculture: Peel the scales at the base of the test tube plantlet and inoculate it on the subculture medium. The formula for subculture for 2 to 5 generations is: MS+0.2~0.3mg./L 6-BA, 3% sucrose; Adjust the formula appropriately according to the germination situation, use 0.2mg./L6-BA for more germination, and 0.3mg./L 6-BA for less germination; the formula for subculture for 5-10 generations is: MS+0.15~0.2mg. /L 6-BA, sucrose 3%; subculture 10-15 generations formula: MS+0.1~0.15mg./L 6-BA, sucrose 3%;
[0025] 3) Rooting culture: In order to prevent seedling degradation and variation, the test-tube seedlings cultured for 1-2 years must be rooted to ensure seedling quality. Cut a single bud and inoculate it on the rooting medium. The formula of the rooting medium is: MS+0.1mg./L NAA, sucrose 6%-8%;
[0026] 4) Low temperature treatment: Lily test-tube seedlings cannot be directly tempered. They must be treated at low temperature to germinate new leaves. The method of low temperature treatment is to put the rooted seedlings in a cold storage (2~3℃) for 45~50 days.
[0027] 5) Smelting seedlings and transplanting: Smelting seedlings should be carried out in a greenhouse. The key to smelting seedlings and transplanting lies in the substrate. It is best to use river sand or sediment for the seedlings. The rooting seedlings were washed to remove the medium and transplanted, with a spacing of 3cm×4cm. Using this substrate to raise seedlings has the advantages of low cost and fewer pests compared with other substrates such as peat.
