According to the method, by simulation of a living environment of the denitrifying phosphorus accumulating bacteria, anaerobic-anoxic two-phase alternate conditions are designed, a phosphorus-free culture medium containing an external carbon source at an anaerobic phase and a phosphorus-free culture medium free of external carbon sources at an anoxic phase are adopted respectively for screening, a domestication culture medium is diluted and then coats screened single strains, the screened single strains are respectively dibbled in a high-phosphorous and low-phosphorous solid medium containing 5-bromo-4-chloro-3-indolyl-phosphate ester, stains simultaneously having blue stains are selected in the two culture mediums, the strains are subjected to streak cultivation in a BTB denitrification culture medium, and straining causing the culture medium to become blue are selected.
