Embodiment

[0048] 1. Sample collection

[0049] Use correct medical techniques to collect urine samples (severe hematuria samples cannot be used for determination), and the collected samples should not be left at room temperature for more than 8 hours; if not tested within 8 hours, the samples should be placed in a refrigerator at 2-8°C; if If it needs to be stored or transported for more than 72 hours, it should be frozen at -20°C and avoid repeated freezing and thawing. Return to room temperature before use, shake gently to mix.

[0050] 2. Preparation before experiment

[0051] ① Take a bottle of concentrated lotion and dilute it 15 times with distilled water;

[0052] ② Adjust the temperature of the incubator or water bath to 37°C, and use it after the temperature is stable;

[0053] ③ Fully mix the magnetic particle suspension until there is no visible precipitation.

[0054] 3 Experimental methods

[0055] ① Take out a certain amount of reaction containers (flat-bottomed test ...

Abstract

The invention discloses a kit for detecting the content of urine microalbumin in serum. The kit comprises an anti-reagent A, an anti-reagent B, a magnetic particle reagent, a calibrator, a quality control product and a luminescent substrate solution, wherein the anti-reagent A is an alkaline phosphatase-labeled urine microalbumin derivative; the anti-reagent B is a FITC-labeled urine microalbuminpolyclonal antibody; and the magnetic microparticle reagent is carboxyl magnetic particles coated with a FITC antibody. The invention discloses a preparation method for the kit. The kit combines chemiluminescence with immunomagnetic particles, provides a near-homogeneous reaction system and adopts a one-step reaction mode, so detection performances (such as sensitivity, precision, a detection range, etc.) are greatly improved, and reaction time is greatly shortened (wherein less than 35 min is needed from the beginning of sample feeding to acquisition of detection results, and the reaction time is obviously less than the reaction time of kits of a same kind); high coupling efficiency and firm bonding are realized; and the preparation method is stable in process, and can greatly reduce product cost while improving product performance.

Description

technical field [0001] The invention relates to the technical field of biochemistry, in particular to a kit for measuring the content of urinary microalbumin (ALB) in a human body using an immunocompetitive method and a magnetic particle chemiluminescence method. Background technique [0002] Microalbuminuria is the presence of tiny amounts of albumin in the urine. Albumin is a normal protein in the blood, but only very small amounts are present in the urine under physiological conditions. Microalbuminuria reflects abnormal protein leakage from the kidneys. When a patient has either high blood pressure or diabetes or both (often co-occurring), changes in blood vessels in the kidneys alter the ability of the kidneys to filter proteins, especially albumin, which allows proteins to leak into in urine. Microalbuminuria is an early sign of diabetes affecting the kidneys and is called diabetic nephropathy. [0003] Under normal human metabolism, there is very little albumin in...
