Abstract

The invention discloses a trace and rapid method for determining the content of algae oil. The method comprises the following steps: (1) taking algae liquid and calculating the quantity concentrationof algae cells in the algae liquid; (2) diluting to obtain algae solutions with different algae concentrations; (3) preparing a mother solution; (4) taking 2 test tubes according to each algae concentration; adding 5 to 10mL of the algae liquid and 5 to 10ml of HCl with the concentration of 1mol / L respectively; after sufficiently mixing, adding 0.1mul of the mother solution into each branch tube according to each algae concentration and adding an organic solvent in step (3) into the other branch tube; taking the branch tubes as contrast tubes and sufficiently mixing; (5) observing under a fluorescence microscope by utilizing exciting light with the exciting wavelength of 400 to 600nm, and photographing and recording a single-cell oil accumulation and change condition; (6) calculating an oil volume; (7) calculating the oil content of the algae liquid with a unit volume. By adopting the method disclosed by the invention, the detection is rapid and simple.
