The method comprises the following steps: collecting pittosporum tobira leaves, drying naturally without direct sunshine, crushing, carrying out thermal backflow extraction with a 75% ethanol solution for multiple times, filtering, combining filtrate, and concentrating till no alcohol smell; enriching the extract whichis concentrated till no alcohol smell on a DA-201 type macroporous adsorption resin column, collecting a 14-15BV eluant of 75% ethanol, concentrating till no alcohol smell, and carrying out freeze drying so as to obtain enrichment freeze-dried powder; carrying out ultrasonic dissolution on the enrichment freeze-dried powder with an upper phase and a lower phase of an equal volume, carrying out HSCCC (High-Speed Counter Current Chromatography) separation purification, collecting an eluant component corresponding to pittosporum tobira glycoside A2according to a spectrum, and concentrating and drying, thereby obtaining pittosporum tobira glycoside A2 respectively, wherein the HSCCC adopts a two-phase solvent system consisting of n-butyl alcohol/ethyl acetate/isopropanol and water in a volumeratio of 2:16:3:16.
