Problems solved by technology

[0004] Disadvantages of the prior art: 1. Digestion fragments have different lengths and sizes and need to be screened; 2. Different adapters need to be added twice to the ends of the restriction fragments; 3. Special A-tails and "Y" tails need to be added to the restriction fragments. ” type connector; 4. The steps are relatively cumbersome, with high technical requirements and time-consuming; 5. The cost of sequencing each sample is relatively high

Embodiment Construction

[0036] The present invention will be further described below in combination with specific embodiments and accompanying drawings.

[0037] 2b-RAD is a simplified RAD genotyping method based on type IIB restriction endonuclease, which provides a powerful technique and method for studying population genomic genetics. In this study, we took Chinese Holstein dairy cows as the research object, constructed a herd of Chinese Holstein dairy cows with clinical mastitis and a normal healthy control group, extracted and constructed the whole genome of cows in the herd, and used Bael endonuclease to analyze the whole genome of all cow samples. The DNA was digested to obtain standard digested fragments, and then sequenced and analyzed on the machine. The specific library construction and sequencing process is ( figure 2 ):

[0038] (1) Digestion: ≥200ng of genomic DNA is digested with type IIB restriction endonuclease;

[0039] (2) Adding linkers: the digested products were added to 5 di...

Abstract

The invention relates to a cow mastitis key SNPs locus rs88640083 and 2b-RAD genotyping and analysis method. The method comprises the following steps: constructing a library and sequencing; Bioinformatics analysis: data filtering, digestion sequence extraction, data alignment, SNP typing, genome-wide association analysis; and performing genome-wide association analysis (GWAS) of clinical mastitisphenotypic traits in dairy cows by BayesA model and Logistic regression model. Compared with the prior art, the present invention has the advantages that compared with RADseq, 2b-RAD sequencing has the following advantages: 1. The length of enzyme fragments is uniform and no subsequent screening is needed; 2, that enzyme section does not need to add a Y-shaped link; 3, the steps are simple; 4, thesequence cost of each sample is low; 5, the sequence takes short time. The invention also constructs two genome-wide association analysis models (BayesA and Logistics); and a Chinese Holstein mastitis key SNPs locus and its corresponding gene (SYK) are screened.

Description

technical field [0001] The invention relates to a method for genotyping and analyzing key SNPs loci rs88640083 and 2b-RAD of cow mastitis. Background technique [0002] Restriction site-associated DNA sequencing (RADseq) technology uses restriction endonucleases to digest the genome to generate DNA fragments of a certain size, and then conducts high-throughput RAD markers generated after digestion by constructing a sequencing library. sequencing. Considered one of the most important scientific breakthroughs of the past decade, RADseq enables genome-wide single-nucleotide detection of tens of thousands of genomes at a time with a single, simple, and cost-effective method Polymorphism markers (single nucleotide polymorphism, SNP), thus promoting genomics research. Compared with other sequencing technologies, this technology has the advantages of high throughput, good accuracy, short experimental cycle, high cost performance, and is not limited by the presence or absence of r...
