The method for promoting fertility of distant hybridization generation ofpaddy disclosed by the invention comprises the step of reducing content of ESA1 in receptor paddy or restraining activity of ESA1 in receptor paddy, thereby acquiring a target paddy with fertility higher than that of the receptor paddy and realizing enhancement of paddy fertility. The receptor paddy is the distant hybridization generation of paddy. ESA1 is the protein shown as a sequence 1 in a sequence table. An experiment proves that the maturing rate of the distant hybridization generation of paddy can be increased, the fertility of distant hybridization generation of paddy can be promotedand the obstacle of distant hybridization of wild paddy and cultivated paddy can be overcome after the expression of ESA1 encoding gene in the invention is restrained.
