Embodiment 1

[0017] Embodiment 1: the cultivation of polar snow algae

[0018] (1) Chlamydomonas nivalis, purchased from Culture Collection of Algae and Protozoa (CCAP), and its preservation number is: CCAP11 / 128.

[0019] (2) Cultivate in two stages in the tubular reactor system: A. In the first stage of production, the activated polar snow algae is inserted into the 3N-BBM medium with a pH of 5.5, and the culture temperature is 14.5°C , under the condition of light intensity 2000±200lx, aerated culture for 6 days to obtain the first-stage fermentation culture; under the culture condition of 14.5 ℃, the polar snow algae can double in 28 hours. B. After completing the first stage, transfer the first stage fermentation culture to nutrient-limiting medium (3N-BBM without NaNO 3 ) was induced, and the culture temperature was 14.5°C, and the light intensity was 5000±500lx, and the aeration culture was carried out for 3 days. The color of the polar snow algae changed from green to red, and the...

Embodiment 2

[0021] Example 2: Production of lipid extracts

[0022] (1) thawing frozen polar snow algae cells, and then mixing with methylparaben (0.1%) to obtain a mixture;

[0023] (2) the concentration of polar snow algae cells in the mixture is about 10g / L;

[0024] (3) Put the mixture at 1200 bar (1.2×10 8 N m -2 ) under the pressure of four times of high-pressure homogenization to dissolve the cell wall of polar snow algae, release the fat and water-soluble components of the cytosol, and obtain a lipid extract.

[0025] At the same time, the polar snow algae cells are lysed and encapsulated in a liquid nanoemulsion to prepare a nanoscale polar snow algae cell lysate emulsion. The polar snow algae cell lysate nanoemulsion produced in this way can simultaneously utilize cytoplasmic solubility and insoluble matter.

Embodiment 3

[0026] Embodiment 3: prepare polar snow algae cell lysate liposome with fluidized bed drying method drying lipid extract

[0027] (1) Lipid extract and isomalt are mixed in a mass ratio of 1:1;

[0028] (3) After the lipid extract is mixed with isomalt, it is dried by a fluidized bed at a spray rate of 50 to 60 g / min, and the average drying temperature is 50° C.;

[0029] (4) The residual moisture after drying is not more than 5%, and the liposome of polar snow algae cell lysate is obtained.

Abstract

The invention discloses application of cell lysate of polar snow alga in preparing medicine or cosmetics for preventing, delaying or treating skin aging. According to the present invention, through experiments, the expression level of an anti-senescence gene mRNA is found to be significantly increased after incubation of the cell lysate liposomes of the polar snow alga; in the treatment group withthe cell lysate liposomes of the polar snow alga, the expression level of the anti-oxidation / senescence gene mRNA shows a positive induction effect on H2O2, while the control group is significantly inhibited after H2O2 stress. Therefore, the cell lysate of the polar snow alga of the present invention can delay skin aging, especially the skin aging caused by oxidation reaction or UV, and can be used to prepare medicine or cosmetics for preventing, delaying or treating skin aging.

Description

Technical field: [0001] The invention belongs to the field of medicine and cosmetics, and in particular relates to the application of polar snow algae cell lysates in the preparation of medicines or cosmetics for preventing, delaying or treating skin aging. Background technique: [0002] The skin is the first barrier to protect the body, and like other tissues, the skin also undergoes an aging process called skin aging. And if the skin is exposed to ultraviolet radiation or polluted air, it will accelerate the skin aging process. Aging skin is characterized by decreased cell regeneration in the epidermis, decreased regeneration capacity of collagen in the dermis, decreased skin elasticity, and lack of moisture, among others. [0003] Antioxidants, such as retinoic acid, are the main active ingredients in cosmetics currently on the market for the treatment of skin aging. At the same time, plant, algae, and fungal extracts rich in natural antioxidants have also been used in ...
