[0053] 3. Isolation of platelets and preparation of washed platelets

[0054] Take cubital venous blood into 3.2% sodium citrate (1:9) anticoagulant tubes, centrifuge at 150g at 22°C for 10 minutes, absorb the supernatant as platelet rich plasma (PRP), and then centrifuge at 1,000 g at 22°C Centrifuge PRP at g for 15 minutes, discard the supernatant to obtain concentrated platelet aggregates, platelet aggregates with appropriate amount of Tyrode Buffer (137mmol / L NaCl, 2.8mmol / L KCl, 10mmol / L HEPES, 1mmol / L MgCl 2 ,12mmol / L NaHCO 3 ,0.42mmol / L NaH 2 PO 4 ,1mg / ml glucose and 2.5mg / ml BSA, pH 7.4) Gently pipet and wash twice, finally, gently resuspend the concentrated platelet pellet and adjust the number of platelets to 2-3×10 8 pieces / ml. Washed platelets were used in two-color flow cytometry experiments to measure the expression of CEA on the surface of platelet membranes under resting conditions and after activation with different concentrations of thrombin.

Embodiment 1

[0062] Example 1 Expression of CEA on human platelets

[0063] In this example, human washed platelets were used, and the expression of CEA on the surface of platelet membrane was detected by double-color flow cytometry under resting state and after activation with different concentrations of thrombin (0.5U / ml, 1U / ml, 2U / ml). From figure 1 A and figure 1 B shows that CEA is expressed on the surface of platelet membrane. At the same time, after being activated by different concentrations of thrombin (0.5U / ml, 1U / ml, 2U / ml), the level of CEA on the platelet surface was significantly increased, and the level of CEA on the surface of platelet membrane was activated by a moderate concentration of thrombin (1U / mL) Significantly increased, and showed a downward trend after activation at a higher concentration of thrombin (2U / mL) ( figure 1 C and figure 1 D). This example suggests that CEA is a protein expressed on the surface of platelet membranes, and the expression level of CE...

Embodiment 2

[0064] Example 2 Expression of CEA on platelet membrane surface in patients with acute coronary syndrome and patients with normal coronary arteries

[0065] In this example, two-color whole blood flow cytometry was used to detect the expression of CEA on the platelet membrane surface of patients with acute coronary syndrome and patients with normal coronary arteries, and the results were expressed as MFI±SE. The expression level of CEA on platelet membrane surface in ACS group was significantly higher than that in normal coronary artery group [1459±149.8vs 429.8±110; P figure 2 A); The expression level of CEA on platelet membrane surface of AMI group and UA group was significantly higher than that of normal coronary artery group [1504±180.7vs1364±273.3vs 429.8±110; P figure 2 B); the expression level of CEA on platelet membrane surface in STEMI group, NSTEMI group and UA group was significantly higher than that in normal coronary artery group [1445±309.6vs 1562±194.2vs 1364±273....

Abstract

The invention discloses application of blood platelet membrane protein in the diagnosis of acute coronary syndrome. The blood platelet membrane protein is a protein-carcinoembryonic antigenelated celladhesion molecule 5(CEACAM5/CEA) that was first discovered by an inventor to be expressed on blood platelet membrane. According to the blood platelet membrane surface CEA level, patients with the acute coronary syndrome and patients with normal coronary arteries are diagnosed and distinguished, the area under the curve(AUC) of the receiver operating characteristic curve(ROC) is 0.86, and clinicaldiagnostic significance is achieved; and when troponin(cTnI) is combined for diagnosis, the AUC reaches 0.91. The blood platelet membrane protein can be used for diagnosis of the acute coronary syndrome, the sensitivity is high, and the specificity is high.

Description

technical field [0001] The invention relates to the application of platelet membrane protein in the diagnosis of acute coronary syndrome, belonging to the field of biochemistry. Background technique [0002] Acute coronary syndrome (ACS) is a type of acute ischemic heart disease, and its high morbidity and mortality have become the focus of international health attention [1] . Acute coronary syndrome is closely related to vulnerable coronary plaques, and the rupture of unstable plaques in the coronary lumen can lead to platelet activation, thrombosis, and even thrombotic occlusion. Thus, platelets play an important role in the progression, instability, and rupture of coronary plaques [2-4] . Platelet surface membrane proteins are also involved in platelet activation, adhesion and aggregation, which can indirectly mediate the pathogenesis and clinical manifestations of acute coronary syndrome [5,6] . Clinically, acute coronary syndrome is mainly diagnosed through the com...
