Embodiment Construction

[0044] figure 1 A microscope 2 for imaging a sample 4 is schematically shown. The sample 4 is located on a sample carrier 6 and embedded in an embedding medium 8 below a cover slip 10 . The sample 4, the sample carrier 6, the embedding medium 8 and the cover slip 10 are not part of the microscope, but they are a routine presence in the sample imaging process. Microscope 2 images sample 8 along optical axis 18 onto detector 16 via objective 12 and tube lens 14 . In this case, an immersion medium 19 , usually realized in fluid form, is usually located between the end of the objective 12 and the cover glass 10 . The immersion medium increases optical resolution because it brings about an increase in numerical aperture.

[0045] The objective lens 12 is designed for specific conditions of the sample and a specific focus position, and is especially optimized for spherical aberration. This design is not suitable for different conditions (deviating conditions). For adaptation, ...

Abstract

A method for preparing a microscope (2) for imaging a sample (4) is provided. The method includes the following steps: providing the microscope for imaging the sample, wherein the microscope comprises an objective having a motor-adjustable objective correction ring (20) for correcting imaging aberrations; acquiring sample information comprising at least one of the following indications: thickness of a cover glass (10), material of the cover glass (10), sample temperature, sample type, sample location on a sample carrier (6), embedding medium (8) of the sample, or immersion medium (19); imaging the sample using the microscope for generating at least one raw image of the sample with a first setting of the objective correction ring (20); and inputting the at least one raw image and the sample information into a machine algorithm (32) and determining a second setting of the objective correction ring, which reduces the imaging aberrations relative to the first setting, by means of the algorithm on the basis of the raw image and the sample information.

Description

technical field [0001] The invention relates to a method for preparing a microscope for imaging a sample, wherein the microscope comprises an objective having an adjustable, in particular motor adjustable, objective for correction of imaging aberrations Correction ring. The invention also relates to microscopic methods. Background technique [0002] Microscope objectives are designed for optimal imaging under specific optical conditions. As resolution increases, the microscope's optical system becomes increasingly sensitive to deviations from the conditions on which it was designed. Image aberrations increase. Therefore, high-performance objectives, especially immersion objectives, have so-called correction collars which act optically on the objective and correct spherical image aberrations which may be caused, for example, by deviating design conditions caused by. Such deviations may be, for example, varying coverslip thicknesses, deviating sample temperatures or tempe...
