Addressing the stage displacement error of a microscope, multiple images of a common microscope scale are acquired. Hough transform image analysis is used to obtain the horizontal and vertical graduation lines of the scale. Feature analysis and detection are then used to determine the actual center point and the error between the actual center point and the horizontal displacement length and the system settings. Error compensation is then used to achieve precise center positioning and accurate horizontal movement of the stage. This invention eliminates the need for mechanical inspection and manual operation, achieving simple, fast, and stable microscope stage calibration through image analysis.
