Problems solved by technology

[0005] Technical problem: The present invention aims at the present situation of routine cutting and grafting of Rhododendron staghorn with low propagation coefficient and cumbersome and difficult operation, and provides a rapid propagation method of Rhododendron staghorn tissue culture that is not affected by seasons and overcomes the aseptic system in the tissue culture process. Difficult to establish, low multiplication coefficient, weak growth of clustered seedlings, difficult rooting, low survival rate of transplanting and other technical problems, explored a large-scale, low-cost production of Rhododendron staghorn rapid propagation method and optimal medium

Embodiment Construction

[0015] The present invention adopts the rapid propagation method of forming clustered buds from tender shoot tips and stem sections, and the present invention will be further described in detail below in conjunction with examples.

[0016] 1. Preparation of explants

[0017] The material is taken from the young shoots of Rhododendron staghorn that grow robustly and are free from diseases and insect pests in the greenhouse of the Rhododendron Planting Resource Garden of Jiangsu Academy of Agricultural Sciences as explants. The explants were carefully scrubbed with a soft brush dipped in detergent, especially the leaf axils, and then rinsed with tap water for 10-12 hours, and set aside.

[0018] 2. Sterilization of explants

[0019] On the ultra-clean bench in the sterile room, treat it with 70% alcohol for 30 seconds, rinse it once with sterile water; soak the tip of the shoot in 0.1% HgCl2 (mercuric chloride) for 5 minutes, and the section of the stem for 8 minutes. The body...

Abstract

The invention relates to a tissue culture and rapid propagation method of rhododendron latoucheae. A woody plant medium (WPM) is taken as a basic culture medium, and a tissue culture and rapid propagation system of the rhododendron latoucheae is established through the isolated culture of the rhododendron latoucheae by proportioning regulation of different hormones. The method sequentially comprises the following steps of: selecting an explant; disinfecting; performing primary culturing; performing propagation culturing; performing rooting cultivation on a strong seedling; and transplanting to obtain a test tube seedling which can keep the original plant genotype, has a high aesthetic property and is better differentiated, wherein a primary induction culture medium of the explant is WPM+IBA 0.15mg/L+TDZ 0.3mg/L+cane sugar 30g/L and the pH value is 5.6; a propagation culture medium of an adventitious bud is WPM+IBA 0.15mg/L+TDZ 0.3mg/L+cane sugar 30g/L and the pH value is 5.6; and a rooting culture medium of the strong seedling is 1/2WPM+IBA 0.5mg/L+NAA 0.2mg/L+cane sugar 20g/L+activated carbon 0.3g/L and the pH value is 5.6. By the method, the propagation speed of the rhododendron latoucheae is greatly increased; and the method is suitable for industrialized commercial production of the rhododendron latoucheae.

Description

technical field [0001] The invention relates to a rapid propagation method of rhododendron staghorn through tissue culture, and belongs to the field of biotechnology. Background technique [0002] Rhododendron is the largest genus of Rhododendronaceae, containing about 960 species, widely distributed in Europe, Asia, North America, mainly in East Asia and Southeast Asia. China is the center of modern distribution and differentiation of wild Rhododendron germplasm resources in the world and the country with the largest population. There are about 542 species, which are mainly produced in Southwest and South China. [0003] Rhododendron latoucheae, also known as Rhododendron latoucheae, got its name because of its zigzag branches, shaped like antlers. Evergreen shrub or small tree, likes mild climate and acidic soil; blooms from March to April, with large and bright flowers, long flowering period and fragrance. The natural vertical distribution range is wide, and it can grow...
