Embodiment 1

[0020] Embodiment 1: the production of phthalate degrading bacterial agent

[0021] 1. Medium preparation:

[0022] 2) Strain preservation medium (solid, 1L): peptone 10g, yeast extract 5g, sodium chloride 5g, agar 20g, add water to 1L, pH 7.0-7.5;

[0023] 5) Strain activation medium (liquid, 1L): peptone 10g, yeast extract 5g, sodium chloride 5g, add water to 1L, pH 7.0-75;

[0024] 6) Seed medium (liquid, 10L): yeast extract 2g, DBP 1g, K 2 HPO 4 58g, KH 2 PO 4 30g, (NH 4 ) 2 SO 4 20g, MgCl 2 1.6g, CaCl 2 0.2g, Na 2 MoO 4 2H 2 O 0.024g, FeCl 3 0.018g, MnCl 2 2H 2 0.015g, pH=6.8-75;

[0025] 7) Fermentation medium (liquid, 200L): K 2 HPO 4 600g, KH 2 PO 4 300g, (NH 4 ) 2 SO 4 200g, MgCl 2 16g, CaCl 2 2g, Na 2 MoO 4 2H 2 O 0.24g, FeCl 3 0.18g, MnCl 2 2H 2 O 0.15g, yeast extract 200g, molasses 300g, add water to 200L, pH 6.8-75;

[0026] All the above media were sterilized at 121°C for 20 min.

Embodiment 2

[0032] Embodiment 2: the degradation effect of phthalate degrading bacterial agent to dibutyl phthalate

[0033] 1) Inoculate 1 mL of bacterial agent in 100 mL of inorganic salt medium containing 500 mg / L dibutyl phthalate; the one without inoculation is used as a control

[0034] 2) After 12 hours, sampling and extraction were carried out, and the content of dibutyl phthalate was measured by high performance liquid chromatography. As a result, it was found that the inoculation group could not detect the existence of dibutyl phthalate, and the degradation rate was about 100%, while the control There was little change in the content of dibutyl phthalate in the group.

[0035] The phthalate-degrading bacterial strain provided by the present invention is preserved in the General Microorganism Center of China Microbial Strain Preservation Management Committee, preservation date: November 16, 2010, classification name: Diaphorobacter sp., preservation number: CGMCCNo.4345, Preserv...

Abstract

The invention provides a bacterial strain which is separated from activated sludge and capable of effectively degrading phthalate compounds, and a production method of the degradation bacteria, belonging to the technical field of biology. Proved by appraisal, the strain is Diaphorobactersp., is preserved in the CGMCC (China General Microbiological Culture Collection Center) and has a preservation number of CGMCC No: 4345; and a 16S rDNA gene sequence of the strain has been submitted to a GenBank database and has a accession number of HQ588349. The production method of the strain comprises the following five steps of: culture medium preparation, strain activation, liquid seed preparation, liquid fermentation and bactericide preparation. The phthalate degradation bactericide produced by utilizing the technical scheme disclosed by the invention can be used for effectively degrading common environmental hormone, namely the phthalate compounds in the environment.

Description

Technical field: [0001] The invention belongs to the field of biological technology, and relates to a bacterial strain (Diaphorobacter sp. QH-6) isolated from activated sludge capable of efficiently degrading environmental hormone phthalate compounds, and a production method thereof. technical background: [0002] Phthalates, also known as phthalates, is a general term for about 30 compounds, which are composed of phthalic acid and corresponding alcohols. They are generally colorless oily viscous liquids, which are difficult to dissolve in water and easily soluble Based on organic solvents, it is not easy to volatilize at room temperature, has low cost, many varieties, and large output. It is often used as a plasticizer to improve the plasticity of plastics. The plasticizers currently used are mainly phthalates. The most widely used phthalate plasticizer is diisooctyl phthalate (DEHP), followed by phthalate diester. Commonly used varieties in my country are: dioctyl phthal...
