Embodiment Construction

[0022] Primers were designed for the FATP-1 gene of intramuscular fat deposition with the above characteristics, and PCR amplification was performed to detect the tissue expression profile of FATP-1 gene in seven tissues of pig heart, liver, spleen, lung, kidney, muscle and fat. The results showed that The expression of FATP-1 gene is the highest in adipose tissue, preliminarily indicating that FATP-1 gene regulates the function of fat deposition in adipocytes.

[0024] 1. Extraction of total RNA from muscle tissue

[0025] Take 50kg of adult Banna mini-pig muscle tissue and immediately put it into liquid nitrogen and save it for later use. Total RNA was extracted from Banna minipig muscle tissue using RNA extraction reagents, and the specific operation was carried out according to the instructions of the kit. The extracted RNA was reverse-transcribed into cDNA and stored at -20°C until use.

Abstract

The invention discloses an intramuscular fat deposition fatty acid transport protein 1 (FATP-1) gene. The gene is the fatty acid transport protein 1 (FATP-1) gene which is separated and cloned from a Bannan miniature pig muscle tissue and is associated with intramuscular fat deposition. The full length of a CDS sequence of the gene is 1,941bp and the CDS nucleotide sequence is shown as SEQ. ID. No. 1. The CDS nucleotide sequence encodes 646 amino acids, and an amino sequence is shown as SEQ. ID. No. 2. The tissue expression spectrums of the FATP-1 gene in seven tissues of a pig, i.e. the heart, the liver, the spleen, the lung, the kidney, the muscle and the fat, are detected, and the result shows that the expression amount of the FATP-1 gene is the highest in fat tissues. The result primarily shows that the FATP-1 gene has a function of regulating and controlling fat deposition in a fat cell.

Description

technical field [0001] The invention relates to the genetic engineering technology of animal molecular breeding, in particular to a fatty acid transport protein 1 (fatty acid transport protein 1, FATP-1) gene technology related to pig intramuscular fat deposition. Background technique [0002] In recent years, due to the excessive pursuit of high lean meat percentage in pig breeding work, the quality of pork has seriously declined. Intramuscular fat (IMF) is an important factor affecting meat quality, which is closely related to pork tenderness, flavor and nutrition, and has been more and more recognized and valued. The deposition of intramuscular fat is a complex physiological and biochemical process, including the synthesis, decomposition and transport of triglycerides in skeletal muscle. Thus, triglyceride metabolism in skeletal muscle underlies intramuscular fat deposition. [0003] Fatty acid transport protein 1 (fatty acid transport protein 1, FATP-1) is a specific t...
