Abstract

The invention discloses a method for quickly culturing verticillium dahliae. The method comprises steps of: firstly, culturing the verticillium dahliae on a potato dextrose agar (PDA) medium, and directly beating the medium to form verticillium dahliae blocks; then utilizing 1ml of sterile water to wash the verticillium dahliae blocks, and obtaining a conidium liquid; finally, sucking the conidium liquid, dipping the conidium liquid onto the PDA medium laid with sterilization filter papers, and culturing at 25 DEG C for 7 days; and scraping to obtain hyphae in a centrifugal pipe of 1.5ml, carrying out liquid nitrogen flash freezing, storing at -80 DEG C for extracting deoxyribose nucleic acid (DNA), and moreover, utilizing the sterile water to wash the PDA medium so as to prepare the conidium liquid. By utilizing the method, a purpose of simply, easily and quickly culturing the verticillium dahliae is realized, and the hyphae and conidia for DNA extraction and inoculation experiments can be greatly cultured within a short time, so that the method has strong practicability and strong popularization and application values.
