Embodiment 1

[0019] (1) Soak animal skin tissue in a solution with an SDS concentration of 0.0087M / L and a NaOH concentration of 0.01M / L at 37°C for 24 hours to obtain an acellular dermal matrix (ADM);

[0020] (2) Put the bFGF DNA in the liposome package, and then put it together with the acellular dermal matrix (ADM) in step (1), and make the bFGF DNA attach to the acellular dermal matrix (ADM) through electrostatic adsorption to form ADM-bFGF complex.

Embodiment 2

[0022] (1) Soak animal skin tissue in a solution with an SDS concentration of 0.0087M / L and a NaOH concentration of 0.01M / L at 25°C for 12 hours to obtain an acellular dermal matrix (ADM);

[0023] (2) Put the bFGF DNA in the liposome package, and then put it together with the acellular dermal matrix (ADM) in step (1), and make the bFGF DNA attach to the acellular dermal matrix (ADM) through electrostatic adsorption to form ADM-bFGF complex.

Embodiment 3

[0025] (1) Soak animal skin tissue in a solution with a concentration of SDS of 1M / L and a concentration of NaOH of 1M / L at 37°C for 24 hours to obtain acellular dermal matrix (ADM);

[0026] (2) Put the bFGF DNA in the liposome package, and then put it together with the acellular dermal matrix (ADM) in step (1), and make the bFGF DNA attach to the acellular dermal matrix (ADM) through electrostatic adsorption to form ADM-bFGF complex.

Abstract

The invention relates to a technical field of decellularization of animal skin tissues in biomedical engineering, and specifically provides an ADM-bFGF (acellular dermal matrix-base fibroblast growth factor) complex and application thereof. The method is as below: treating the animal skin tissues in a solution containing SDS (sodium dodecylsulfate) and NaOH to obtain ADM; and wrapping the bFGF DNA into liposome, placing the bFGF DNA with ADM, adhering the bFGF DNA on the acellular dermal matrix through electrostatic adsorption, so as to form an ADM-bFGF complex. Through a large amount of verifications, the acellular dermal matrix is suitable for covering of wounds, granulation wounds, chronic ulcers and bedsore, and playing the effects of preventing water evaporation, loss of protein and heat, and microbial infection, and promoting wound healing. The acellular dermal matrix has good biocompatibility, no immunogenicity or low immunogenicity, no toxicity and no allergy, and is safe and easy to use.

Description

technical field [0001] The invention relates to the technical field of decellularization of animal skin tissue in biomedical engineering, in particular to an acellular dermal matrix-basic fibroblast growth factor complex (ADM-bFGF complex) and its application. Background technique [0002] With the development of society and economy in various countries in the world, the improvement of residents' living standards, the changes in diet structure, the increasingly tense pace of life, and the lifestyle of less movement and more sitting, the global incidence and prevalence of diabetes are increasing rapidly. Diabetes It has become the third chronic disease that seriously threatens human health after tumors and cardiovascular diseases. The skin of diabetic patients is easily damaged, and after the injury, it often recurs and persists, forming stubborn and difficult-to-heal ulcers. However, since the mechanism of the refractory wounds caused by diabetes is still unclear, conventio...
