Embodiment 1

[0025] Embodiment 1 Obtaining of Raman spectrum peaks of the present invention

[0026] 1 Experimental equipment and materials

[0027] 1.1 Experimental equipment

[0028] Laser Raman spectroscopy system: dispersive confocal Raman spectrometer (Senterra R200-L), produced by Bruker, Germany, with spectral resolution ≤ 1.5cm -1 , horizontal resolution <1μm, vertical resolution <2μm, equipped with three kinds of lasers with wavelengths of 785nm, 633nm and 532nm respectively, three-dimensional automatic control platform and other important performances; Spanish CASA analyzer; Eppendorf 5424 small high-speed centrifuge.

[0029] 1.2 Experimental materials

[0030] 10 semen samples from donor volunteers in the Shanghai Human Sperm Bank of Renji Hospital Affiliated to Shanghai Jiaotong University School of Medicine were selected, and the 10 sperm samples were all fertile male volunteers. Put 4 eggs into each semen sample and incubate in a 37°C incubator for 2 hours. From each s...

Embodiment 2

[0062] Example 2 The Raman spectral peaks of the present invention can be used to judge the verification of sperm fertilization function

[0063] 1 Sample source

[0064] A total of 60 patients were collected in the clinical andrology outpatient clinic. The subjects were informed of the purpose of the experiment and signed the informed consent. 10 sperm were picked from each subject's semen. A total of 600 test sperm were randomly divided into 3 200 spermatozoa were tested in each group.

[0065] 2 Laser Raman microscope scanning

[0066] First adopt the Raman spectrometer to scan the acrosome region of the tested sperm according to the method of Example 1, obtain the Raman spectrum peaks of the anterior region and the rear region of the sperm acrosome, and then use the Raman spectrum peaks of the obtained Raman spectrum with the Raman spectrum of the present invention The peaks were compared to investigate whether the Raman spectrum peaks in the anterior and posterior reg...

Abstract

The invention relates to a sperm acrosome zone Raman spectrum peak and a use thereof. The Raman spectrum peak is represented by an abstract figure shown in the specification, and can be used for the fertilization function assessment and detection and the theory researches of a sperm. The invention also relates to a sperm fertilization function assessment method. The method comprises the following steps: 1, preparing a spermatic slice; 2, scanning the single sperm acrosome zone on the spermatic slice at a laser excitation wave of 532nm under a laser power of 5mW three times to obtain a Raman data spectrum peak; and 3, contrasting the Raman data spectrum peak of a detection sample with the Raman data spectrum peak of the above sperm acrosome zone to determine that whether the sperm has a fertilization function or not. The method provides a standard Raman spectrum peak for determining the sperm fertilization potential, and the determination result of the method is accurate and reliable, and has a high credibility, so the method is a new sperm fertilization potential detection method; and a separate sperm can be controlled by the method, so there is no need to specially process the sperm, the damage of the sperm can controlled in an extremely low range, and the sperm can be directly used for subsequent researches or auxiliary reproduction treatment.

Description

technical field [0001] The invention relates to the technical field of sperm fertilization function detection, in particular to a Raman spectral peak in the acrosome region of sperm and its application in detecting sperm fertilization function. Background technique [0002] The evaluation and detection of sperm fertilization function is an important means for the diagnosis and treatment of male infertility, the establishment of male contraceptive technology, the artificial breeding of rare animals, and the cultivation of superior species of economic animals. In-depth study of the structure and function of human sperm is of great significance for exploring the mystery of spermatogenesis and giving birth to healthy offspring. [0003] Sperm is an important biomolecule in living organisms. Sperm consist of a complex mixture of organic, inorganic, and biochemical substances contained within cell membranes, including nucleic acids, proteins, polysaccharides, and fluids. The com...
