Problems solved by technology

[0004] There are not many reports on the extraction methods of octopus polysaccharides, mainly including cold water extraction and enzymatic extraction. Generally, column chromatography is used for purification, which is relatively cumbersome.
[0005] Most of the products in my country's octopus processing industry are exported, and the products are mainly octopus meat processed food. However, a large amount of waste water is generated during the processing process, which has not been effectively used yet.
However, there are no relevant studies and reports on the extraction of octopus polysaccharides from octopus processing wastewater.

Embodiment 1

[0026] Embodiment 1, a kind of method utilizing octopus processing waste water to extract polysaccharide, carries out following steps successively:

[0027] 1) Concentrate 1000L of octopus processing wastewater at 55-65°C to 1 / 20 of its original volume;

[0028] 2) Add papain with a mass volume ratio of 1% to the concentrated solution obtained in step 1) (10g papain / 1000ml concentrated solution), adjust the pH to 7.0, hydrolyze at 60°C for 6 hours, and heat up to 90°C Insulated for 10 minutes to obtain hydrolyzate;

[0029] 3) Add 4 times the volume of 95% (volume %) ethanol to the hydrolyzate obtained in step 2), stir, and place at 4°C for 12h (that is, overnight in a refrigerator at 4°C); centrifuge at 5000 rpm for 10min, and collect the polysaccharide precipitate;

[0030] 4) Take the polysaccharide precipitate obtained in step 3), add 10 mass volume times (1g / 10ml) distilled water to dissolve; add Sevag reagent (chloroform: n-butanol = 4:1 volume ratio) that is 4 times th...

Abstract

The invention discloses a method for extracting polysaccharide through octopus processing wastewater. The method comprises the following steps: concentrating octopus processing wastewater and hydrolyzing the octopus processing wastewater through papain or trypsin, adding ethanol to the obtained hydrolysate and stirring, adding distilled water to an obtained polysaccharide precipitate, adding a Sevag reagent, and centrifuging; adding ethanol to an obtained solution to stir and centrifuge, washing the collected polysaccharide precipitate, and freeze-drying under a vacuum condition to obtain crude octopus polysaccharide; and adding distilled water to the crude octopus polysaccharide to obtain crude polysaccharide liquid, filtering the crude polysaccharide liquid through an ultra-filtration membrane which is 300K in molecular weight cutoff and an ultra-filtration membrane which is 100K in molecular weight cutoff sequentially, and freeze-drying an obtained material under a vacuum condition to obtain octopus polysaccharide which is 300-100K in molecular weight. The method disclosed by the invention is used for extracting polysaccharide in the octopus wastewater for the first time; therefore, the method has a technical advantage of turning waste materials into things of value.

Description

technical field [0001] The invention relates to a method for extracting polysaccharides, in particular to a method for extracting polysaccharides by using octopus processing wastewater. Background technique [0002] As an important marine economic aquatic product, octopus has been favored by consumers as a high-end aquatic product because of its thick meat, rich nutrition, delicious taste, unique taste, and high medical value. It is rich in a large number of extremely valuable natural bioactive substances: taurine, octopus carnitine, collagen, octopus toxin, etc., which play a role in regulating blood pressure, treating neurasthenia, antibacterial disinfection, promoting digestion, enhancing memory, and preventing dementia. It has an important role and can be used as a clinical drug and a high value-added health food additive. [0003] Previous studies believed that polysaccharides are important functional components contained in marine animals, and have anti-tumor, immune-...
