Embodiment 1

[0034] The use of embodiment 1Miliflex Quantum kit

[0035] Mimiflex Quantum kit includes: 1 Reconstitution solvent, 2 Fluorescent agent, 3 Staining buffer.

[0036] Remove the Miflex Quantum kit from the refrigerator 1 hour before starting reconstitution. Reconstitute under a laminar flow hood or under a microbiological safety cabinet. Shake bottle 1 at room temperature until the reconstitution agent is completely dissolved, transfer the reconstitution agent in bottle 1 to the fluorescent agent in bottle 2, and mix well. Transfer the well-mixed reagent in bottle 2 to the staining buffer in bottle 3, mix well, and use it as the staining solution.

[0037] Take out the Milliflex culture plate in the incubator and put it into a laminar flow table or a microbial safety cabinet; wipe the surface of the membrane transfer device with 1 piece of non-woven cloth moistened with sterile denatured ethanol 70 / 30; put the Milliflex liquid culture medium The plate is placed on the membra...

Embodiment 2

[0044] The microorganism that embodiment 2 detects

[0045] The types of microorganisms detected include: spirulina spirulina, Chryseobacterium spp., Copper greedy spp., Ralstonia pieterii, Sphingomonas spinosa, Caulobacter spp., Acinetobacter spp., Hemoglobin Alcoholomonas, Acidovorax and Corynebacterium fischeri.

[0046] As a control, the traditional membrane filtration method was used.

[0047] The parameters of the two methods are compared in Table 1.

Abstract

A method for quickly detecting microbes in pharmaceutical water comprises the steps as follows: (1) filtering the to-be-detected pharmaceutical water, transferring a Milliflex membrane to a plate containing a culture medium; (2) transferring the Milliflex membrane cultured for a short period to a plate containing a staining solution; (3) performing staining for 30 min with a staining fluid prepared by a staining kit; (4) counting microbe bacterial colonies which emit fluorescence through an observation device, and recording the count as FC; (5) transferring the Milliflex membrane to the plate containing the culture medium for culture for 5 days, recording the count as RIC, counting conventional bacterial colonies for the Milliflex membrane cultured for 5 days, and recording the count as CC; (6) studying the FC at different time points, and comparing RIC (reduced-intensity conditioning) results with the CC. With the adoption of the method, whether the pharmaceutical water is polluted or not can be determined more quickly, the production interruption of products can be avoided, and microbe experiment results can be obtained earlier, so that better understanding and mastering for production water can be realized. The method is applicable to microbiological labs and pharmaceutical factories.

Description

technical field [0001] The invention relates to a method for detecting microorganisms, in particular to a method for rapidly detecting microorganisms in pharmaceutical water. Background technique [0002] The rapid detection method of microorganisms is a rapidly developing direction in microbiology. Whether it is the supervision and management of food and pharmaceutical water safety, a relatively rapid microbial detection method is required. [0003] At present, for the detection of microbial bacteria content, the traditional method is mainly used to inoculate the object to be detected into a suitable medium, culture, observe and count. [0004] However, the current method needs several days due to the need for long-term cultivation, which is unfavorable for the timely detection of the bacteria content in the object to be tested. Contents of the invention [0005] The object of the present invention is to provide a method for rapidly detecting microorganisms in pharmaceut...
