Embodiment 1

[0067] Embodiment 1, adopting baicalein soaking treatment before irradiation treatment

[0068] 1. Take biomaterials rich in collagen tissue

[0069] 1. Collect the esophageal tissues of adult pigs that have just been slaughtered from slaughterhouses under standardized management, try to avoid contact with pollutants, and freeze them immediately after collection.

[0070] 2. Thaw the esophageal tissue obtained in step 1 and wash it thoroughly, and remove the part that is not suitable for processing (strip the muscle tissue, take the part with a length of about 15-20 cm in the middle and a uniform diameter).

[0071] 2. Preparation of collagen materials for bioremediation

[0072] 1. The first alkali treatment

[0073] Use alkaline solution to soak the product of step one.

[0074] The purpose of this step is to destroy the cell structure, destroy and dissolve non-collagen components.

[0075] Specific steps: use 1M NaOH aqueous solution, soak at 6±2°C for 1 hour, and wash ...

Embodiment 2

[0153] Embodiment 2, adopting baicalin soaking treatment before irradiation treatment

[0154] 1. Take biomaterials rich in collagen tissue

[0155] Same as Step 1 of Example 1.

[0156] 2. Preparation of collagen materials for bioremediation

[0157] Except step 4, others are the same as step 2 of embodiment 1.

[0158] 4. Radiation protection agent treatment

[0159] Specific steps: soak in 0.01g / 100mL baicalin solution at 6±2°C for 6 hours, and wash with water.

[0160] Preparation method of baicalin solution: take 0.01g baicalin, dilute to 100ml with water, then add 0.07g NaOH, stir for about 10min, baicalin is fully dissolved.

[0161] It is set to carry out the comparison processing of the above-mentioned step 1, step 2, step 3, step 5 and step 6 continuously.

[0162] 3. Preparation of collagen materials for bioremediation

[0163] Except step 4, others are the same as step 3 of embodiment 1.

Embodiment 3

[0186] Embodiment 3, adopt apigenin soaking process before irradiation treatment

[0187] 1. Take biomaterials rich in collagen tissue

[0188] Same as Step 1 of Example 1.

[0189] 2. Preparation of collagen materials for bioremediation

[0190] Except step 4, others are the same as step 2 of embodiment 1.

[0191] 4. Radiation protection agent treatment

[0192] Specific steps: soak in 0.3g / 100mL apigenin solution at 14±2°C for 20 hours, and wash with water.

[0193] Preparation method of apigenin solution: take 0.3g apigenin, dilute to 100ml with water, then add 0.02g NaOH, stir for about 10min, apigenin is fully dissolved.

Abstract

The invention discloses a method for protecting a biological material before irradiation sterilization. According to the method provided by the invention, 0.01-0.5g/100ml of an irradiation protective-agent solution is used for immersing and processing the biological material at 0-25 DEG C for 1-24 hours, wherein the irradiation protective-agent solution is a natural flavones solution or flavonoid solution. The selected flavonoid reagent is used as a stable and effective irradiation protective agent for irradiation protection of the product so as to reduce the damage of the irradiation process to an inherent protein structure of the product to the maximum. The protective effect is obvious and performance is stable; residues are easy to control and have low toxicity; and the product will not be damaged and the environment will not be polluted.

Description

technical field [0001] The invention relates to a method for protecting biological materials before irradiation sterilization. Background technique [0002] In modern surgery, natural collagen scaffolds are often used to repair some defective tissues and organs. The main component of natural collagen scaffold materials is collagen, which needs to be sterilized before use. Conventional sterilization methods are physical sterilization and chemical sterilization. [0003] Physical sterilization can be divided into moist heat sterilization and dry heat sterilization, both of which require high-temperature treatment of collagen scaffold materials, which will cause collagen inactivation during high-temperature treatment. [0004] Chemical sterilization usually selects a reagent or gas with a sterilizing effect to sterilize the collagen scaffold material. Common reagents include ethylene oxide, lactic acid, etc., which are biologically toxic, and it is difficult to control their...
