Problems solved by technology

At present, spraying fungicides and promoting disease-resistant varieties are the main control methods in production, but long-term use of drugs will cause environmental pollution and physiological race variation of pathogens, so promoting disease-resistant varieties is the safest and most effective method
However, there are few literature reports on the study of cabbage powdery mildew, which seriously hinders the pace of research on cabbage powdery mildew.

Embodiment Construction

[0031] The identification of disease resistance genes plays an important role in the research of crop resistance genetic theory and the selection of disease resistance varieties. This method can quickly identify the cabbage powdery mildew gene. The specific implementation process is as follows:

[0032] 1) Collection and identification of cabbage MLO gene

[0033] In order to obtain all the members of the MLO type gene family of cabbage, we first constructed the HMM model based on the MLO type genes of Arabidopsis thaliana, and the powdery mildew resistance MLO gene sequences of tomato, pea, pepper, rose, pepper, and Lotus root. In the collection of MLO type genes; secondly, the published MLO gene sequences in different crops were used as target sequences (from DFCI database: TC171015, TC267529, DFCI: TC327983, TC289653, TC312087, TC132500, TC133436, TC317623, TC317025, TC315947, TC325903, TC315944, TC315912, TC322759, TC322059, TC330654, TC282713, TC293173, TC281861, TC283253, T...

Abstract

The invention provides a rapid identification method of powdery mildew-resistant genes of cabbage, relating to subject knowledge such as comparative genomics of plants, genetics and bioinformatics and belonging to the field of plant biologic technical science. The rapid identification method mainly comprises the following steps: (1) downloading a whole genome sequence of the cabbage, and collecting MLO type genes; (2) identifying the MLO type genes; (3) establishing phylogenetic relationship of the MLO type genes; and (4) comparing MLO type powdery mildew genes. According to the rapid identification method, the mining period of powdery mildew genes of the cabbage is effectively shortened, and the rapid identification of the powdery mildew genes is benefited; by developing corresponding coseparation functional markers (SNP, SCAP and the like) by virtue of identified candidate powdery mildew genes, the molecular marker-assisted selection of the powdery mildew-resistant genes can be rapidly carried out, and the accuracy is high; by combining molecular markers of other disease-resistant genes, multi-resistance breeding materials can be created, the breeding age limit can be shortened, and the breeding efficiency can be improved; a foundation is laid for the exposition of a powdery mildew-resistant molecular mechanism of the cabbage.

Description

Technical field [0001] The present invention is to quickly identify cabbage powdery mildew genes by means of sequencing the whole genome sequence of cabbage, using plant comparative genomics, genetics, bioinformatics and candidate gene strategies, etc. It mainly involves the download of cabbage whole genome sequence and the identification of candidate genes , Gene comparison, clustering and other means to identify powdery mildew genes, which belong to the field of plant biotechnology science. Background technique [0002] Cabbage belongs to the Brassica genus of the cruciferous family. It is an annual or biennial herb and is one of the important vegetables. It is rich in vitamins, sugar and other ingredients, of which vitamin A is the most, and contains a small amount of K, U, chlorine, iodine and other ingredients. Eating cabbage can relieve mild ulcers or duodenal ulcers. In addition, some sulfides contained also have anti-cancer effects. Powdery mildew belongs to the subphy...
