Embodiment Construction

[0021] The technical solutions of the present invention will be further described in detail below in conjunction with the accompanying drawings and specific embodiments.

[0022] Using this method, two large-scale experiments have been carried out in March-June 2014 and August-November 2014: Experimental results:

[0023] According to this method, the first experiment (March-June, 2014) processed 1921 Newhall navel orange plant axillary buds (67 plants); the second experiment (August-November, 2014) processed 3088 Axillary buds of Newhall navel orange plants (125 plants). Observing the phenotype of the young leaves of the plants after the treatment, it was found that compared with the normal state where the leaves of Newhall navel orange were oval, thick and dark green, and relatively flat, the morphology of the new leaves of some plants after treatment had obvious variation. Some of the leaves on the branches developed from some axillary buds have irregular leaf margins, som...

Abstract

The invention discloses a handling method for inducing citrus bud mutation through NaN3. The method includes the following steps that NaN3 conditioning fluid is prepared; citrus one-to-two-year-old grafted seedlings are selected, the part, which is not lignified completely, of a branch to be handled is cut off, and all leaves and germinal buds of the reserved part of the branch are removed; all leaf axils of the reserved part of the branch are incised by a sharp blade; the branch to be handled is wrapped with absorbent paper by one to two layers, and the wrapped absorbent paper is wetted by the NaN3 conditioning fluid to be saturated through a liquid-moving machine and the like; the branch is covered with a preservative film until handling is completed, so that the conditioning fluid is prevented from being evaporated; the wrapped absorbent paper is uncovered, a plant is placed under normal culture conditions such as the outside of a room to grow, it can be observed that new shoots can germinate and grow from certain leaf axils after five days, and afterwards, variation character observation and statistics of leaf morphology and the like can be conducted. In the culture growth process after handling, the buds growing at the unhandled part of the plant need to be removed in time, in accordance with experimental result statistics, the mutagenesis effect is obvious, and application value can be achieved.

Description

technical field [0001] The invention belongs to the technical field of biotechnology research and development, and relates to a treatment method for sodium azide-induced bud change of citrus. Background technique [0002] Compared with other chemical mutagens, sodium azide (NaN 3 ) has the advantages of no residual toxicity and relatively safe use, but so far there has been no research report on the method of using sodium azide to mutate citrus axillary buds. Sodium azide has been applied in the breeding research of plants such as barley, rice, sunflower, soybean, flower and apple, and some beneficial mutated plant materials have been obtained; There are following defects in the processing method of inducing citrus bud change: [0003] 1. The concentration (reagent preparation) and treatment time of sodium azide to effectively trigger citrus budding are not clear [0004] So far, there is no research report on how to treat citrus plants with sodium azide to effectively ca...
