Abstract

The invention relates to the field of medicine, in particular to application of a tilia amurensis inflorescence extractive to preparation of a medicine for treating insomnia. A preparation method of the tilia amurensis inflorescence extractive comprises the following steps: extracting tilia amurensis inflorescence by using 70% ethyl alcohol, then extracting the tilia amurensis inflorescence by using ethyl acetate to prepare a crude extractive, carrying out gradient elution on the crude extractive through a dichloromethane-methyl alcohol mixed solution by using a silica gel column chromatography, collecting eluant prepared by the dichloromethane-methyl alcohol mixed solution with dichloromethane and methyl alcohol in a volume ratio of 15 to 1, and recycling a solvent to prepare a purified extractive, wherein the purified extractive of the tilia amurensis inflorescence is separated by an ODS column chromatography and then is eluted by a 50% methanol aqueous solution to obtain two monomeric compounds; by virtue of NMR identification, the two monomeric compounds are tiliroside and astragalin; the tiliroside is an active component; the purified extracted is analyzed by HPLC; the mass content of the tiliroside is 31.7%. Animal experiments show that the tilia amurensis inflorescence extractive is obvious in resistance to the insomnia; the dose-effect relationship research shows that the tilia amurensis inflorescence extractive is free of obvious dose dependence and low in addiction possibility; the safety of the tilia amurensis inflorescence extractive is superior to that of commonly-available chemical medicines at present.
