[0020] Microsporidia universal primers (V1F / 1492R) and nested primers (MI1F / MI545R and MI230F / MI426R) were used to perform PCR amplification experiments on the muscle DNA templates of 76 wild Portunus trituberculatus samples.

The amplification conditions were: 94°C for 5 min, followed by 32 cycles of 94°C for 45 s, 45°C for 35 s, and 72°C for 90 s, and finally 72°C for 10 min. The PCR products were subjected to 1% agarose gel electrophoresis, and the results were observed with an ultraviolet gel imager.
