The invention discloses a preparation method of a Macrochelys temminckii extract. The preparation method comprises the following steps: 1) collecting blood after killing Macrochelys temminckii, removing viscera and shell, and retaining liver, gallbladder and meat and dicing; and mincing Macrochelys temminckii meat dices, liver and gallbladder by the use of a mincing machine; (2) pouring the minced Macrochelys temminckii meat, liver and gallbladder and the collected Macrochelys temminckii blood into a reaction kettle for a reaction, and adding protease and flavour enzyme to carry out enzymolysis; and killing enzyme after enzymolysis; 3) grinding a solid-liquid mixture obtained after enzymolysis into a paste material through a three-stage colloid mill, pumping the paste material through a material suction pump into another reaction kettle and adding maltodextrin, an emulsifier and TBHQ, stirring for 10 min, sieving the paste material through a 40-mesh vibrating screen, and emulsifying the sieved paste material for 15 min after pumping the sieved paste material into an emulsifying kettle; 4) sending an emulsion obtained after emulsification into a homogenizer and homogenizing under the condition of 20-50 MPA, pumping a homogenized material into an interlayer storage tank, storing and heating to 90-100 DEG C and sterilizing for 30 min; and 5) making the homogenized material in the storage tank into a powder through a spray drying tower; and naturally cooling and carrying out sealed packaging by an automatic packaging machine.
