Problems solved by technology

[0004] For many years, the control of wheat head blight has mainly relied on chemical pesticides. However, with the environmental problems caused by chemical pesticides and the emergence of drug-resistant Fusarium graminearum strains caused by the use of chemical pesticides for many years, the control has become more difficult.
Although a lot of energy has been invested in related research at home and abroad on the quality of scab resistance, there is still no ideal high-resistant wheat variety (Li Zhenghui et al., 2007).

Embodiment 1

[0031] Isolation and Identification of Example 1 Bacterial Strain JS25R

[0032] 1. Isolation of bacteria

[0033] (1) In the ultra-clean workbench, 30 ears of wheat diseased ears collected in Jiangsu (June 2012) were shaken in 100 ml of sterile distilled water for 15 minutes to prepare a bacterial suspension, and then treated in a water bath at 80°C for 10 minutes.

[0034] (2) Dilute the bacterial suspension in a concentration gradient with sterile distilled water, spread it on the NA medium plate, and cultivate it at 30°C for 24 hours. The colonies cover the entire plate. Use an inoculation loop to pick out the shape, size, The strains with different colors and transparency were streaked and purified on the plate, and the purified strains were applied to the confrontation culture experiment of Fusarium graminearum. The obtained strain with strong antagonistic ability to Fusarium graminearum was named JS25R.

[0035] 2. Identification

[0036] (1) According to the method ...

Embodiment 2

[0046] Antagonism of Example 2 Bacillus velei JS25R to Fusarium graminearum

[0047] The antagonism of Bacillus velei JS25R to Fusarium graminearum was detected by confrontation culture method.

[0048] 1. Use a hole puncher with a diameter of 5 mm to pick out the bacterial plate at the edge of the cultivated pathogenic fungus Fusarium graminearum (F. graminearum) J07 colony, and pick the bacterial plate into the center of the PDA plate with a sterilized toothpick.

[0049] 2. Inoculate the same volume and concentration of JS25R on a four-dot dash line at a distance of 25mm from the center of the PDA plate. Take the PDA plate inoculated only with the plate of Fusarium graminearum as the control group, culture at 28°C for 4-5 days, and observe whether there is antibacterial effect bands formed. Do three parallels for each group. The results are shown in Table 1 and figure 1 Shown:

Embodiment 3

[0053] The inhibitory effect of embodiment 3 Veles bacillus JS25R fermented liquid to Fusarium graminearum

[0054] The antagonism of Bacillus velei JS25R to Fusarium graminearum was detected by confrontation culture method.

[0056] 2. Inoculate JS25R with the same volume and concentration at four points 25mm away from the center of the PDA plate. Take the PDA plate inoculated with only the plate of Fusarium graminearum as the control group, culture at 28°C for 4-5 days, and observe whether there is any antibacterial effect. bands formed. Do three parallels for each group. The results are shown in Table 2, indicating that JS25R has a strong inhibitory effect on Fusarium graminearum.

Abstract

The invention discloses Bacillus velezensis JS25R and an application thereof to biological control of plant diseases caused by fusarium. The strain JS25R is separated from diseased wheat ears, the preservation number is CGMCC (China General Microbiological Culture Collection Center) NO.14240, and the strain not only can efficiently inhibit fusarium graminearum when cultivated on a plate, but also has the control effect as high as 77.4% in a greenhouse experiment. As a biological control material for fusarium head blight of wheat and maize, the strain has good application prospect for development of novel biopesticides or biological control microbial agents.

Description

technical field [0001] The invention relates to the technical field of microbiology and plant disease biocontrol, in particular to a strain of Bacillus Velez JS25R and its application. Background technique [0002] Fusarium graminearum (Fusarium graminearum) is one of the important pathogens of gramineous crops, which can cause wheat head blight (Fusarium Head Blight, FHB). Fusarium graminearum is a major factor limiting wheat yield (Dubin, 1997) . my country's climatic conditions are very favorable for the occurrence of wheat head blight, and the perennial disease in the middle and lower reaches of the Yangtze River caused a yield loss of 10%-15%, and a production reduction of nearly 50% in a pandemic year. Due to the impact of global warming and changes in farming systems such as straw returning to the field and no-tillage cultivation, the occurrence area of wheat head blight in my country has rapidly expanded from the Yangtze River Basin to Northwest and North China, e...
