Abstract

The invention discloses a 3D cell culture chip based on soft lithography as well as a preparation method and an application of the 3D cell culture chip. The preparation method comprises steps as follows: a mask provided with a set pattern structure is manufactured with the soft lithography; the mask is uniformly covered with a liquid polymer compound or a polymer compound solution, and a liquid layer with a certain thickness is formed; after the liquid layer is cured, the mask is removed, and a seal with a set 3D structure arranged on the printing surface is obtained; the printing surface is contacted with a modifier, so that the modifier can be attached to the printing surface in a separable manner; the printing surface and the surface of a selected substrate are contacted, then, the sealis moved away from the selected substrate, so that at least part of the modifier is separated from the printing surface and attached to the surface of the selected substrate to form a pattern structure, wherein the pattern structure comprises a plurality of arrays formed by patterns capable of adsorbing single cells. The 3D cell culture chip is easy and simple to prepare on a large scale at a lowcost and can provide an ideal 3D cell culture environment.
