Abstract

The invention relates to a tissue-culture seedling raising method for kadsura coccinea tender stems with buds. The method comprises the technical procedures of explant anti-browning treatment, disinfection treatment, axillary bud induction culture, callus induction culture, cluster bud induction differentiation and rooting culture. The method adopts the freshly germinated tender stems with buds asexplants, and the explants are soaked in antioxidant Vc distilled water solution and then soaked in potassium permanganate distilled water solution; finally perlite and activated carbon with large porosity are added in a three-stage culture medium of initial induction, callus induction and subculture proliferation culture to absorb the white mucus and harmful substances produced after splitting of the kadsura coccinea tender stems, and PVP is added to inhibit the production of browning substances such as phenols and the like. By adopting the method, the initial induction contamination rate and the browning rate can be controlled less than 15%, the value-added coefficient is 4.0 or above, and the induced survival rate and the rooting rate are both 85% or above. The tissue-culture seedlingraising method is systematically reported for the first time, and a reference is provided for system optimization and integration of a kadsura coccinea improved variety factory-like seedling raising technique.
