Embodiment 1

[0038] Weigh a certain amount of high polyselenium powder sample, add 10 times the volume of ultrapure water, place in a constant temperature water bath oscillator at room temperature 25 °C and shake at 200r / min for 1h, mix evenly, after fully extracting, suction filter, take the upper The supernatant was passed through a 5 μm water-based filter membrane, and the extract obtained was stored in cold storage at 4°C.

[0039]Take 2ml of the extract, after measuring its pH value, adjust the pH to 3.5 with a low-concentration acetic acid solution, stir well, and slowly add a precooled saturated ammonium sulfate solution whose volume is 50% of the extract volume in the extract, and lightly add while adding. Stir lightly to prevent the local concentration of ammonium sulfate from being too high, and place it in a refrigerator at 4°C for 1 hour to observe the precipitation of crude protein precipitates. Then place it in a high-speed refrigerated centrifuge (at a temperature of 4°C and...

Abstract

The invention discloses a method for extracting selenium-containing vegetable proteins. The method comprises the following steps: preprocessing Thlaspi arvense; adding an extractant, shaking the Thlaspi arvense and the extractant, performing suction filtration, taking the obtained supernatant, allowing the supernatant to go through a hydrophilic filter membrane in order to obtain an extract, and performing cold storage on the extract; adjusting the pH value of the extract, adding a pre-cooled saturated salt solution while stirring, and standing the obtained solution until crude proteins precipitate; centrifuging the obtained solution in a high-speed refrigerated centrifuge, and removing the obtained supernatant to obtain a lower layer protein precipitate; adding the extractant to re-dissolve the precipitate, and stirring the extractant and the precipitate until the precipitate is completely dissolved in order to obtain a purified protein solution; dialyzing the purified protein solution; transferring out a liquid in a dialysis bag if the dialysis is complete in order to obtain a selenium-containing protein solution; adjusting the pH value of the selenium-containing protein solution, adding the pre-cooled saturated salt solution, and standing the obtained solution until a selenium-containing protein precipitate precipitates; and centrifuging the obtained solution in the refrigerated centrifuge to obtain the selenium-containing vegetable proteins. The method is based on extraction of the vegetable proteins from Thlaspi arvense, so the extraction rate of the vegetable proteinsand the selenium content of the vegetable protein are significantly improved.

Description

technical field [0001] The invention relates to a protein extraction method, in particular to a selenium-containing plant protein extraction method. Background technique [0002] Protein is an essential nutrient for human and animal life, including vegetable protein and animal protein. Selenium is one of the essential trace elements for animals and humans. In the soil-plant-human system, it initially comes from the soil, undergoes plant photosynthesis, and is finally stored in plants in the form of selenoamino acids and selenoproteins, and then enters through the food chain. Therefore, selenoprotein is an important nutrient for human body to supplement selenium. [0003] At present, there are many extraction techniques for plant selenoproteins, including trichloroacetic acid-acetone precipitation (TCA) method, E-TCA (extract solution added EGTA) method, phenol extraction, alkali extraction, acid extraction and water extraction, etc. . To extract protein, the first step is...
