The preparation method of the water plugging agent comprises the following steps of: step 1, introducing nitrogen gas into a reaction container to replace air,then adding the L-lactide, the aspartic acid, the 2-ethyl-2-oxazoline, the dimethyl carbonate into the reaction container, and stirring evenly; step 2, heating a reaction solution to 110-130 DEG C, adding the catalyst and a regulator, adjusting the pH value of the reaction solution to be neutral, heating to 140-160 DEG C, reacting for 8-10 hours at constant temperature, and cooling; step 3, processing the cooled product into particles or powder, namely the water plugging agent.
